Antioxidative effect of velvet antler extracts in vitro and in vivo

摘要 目的以秀丽隐杆线虫为模式生物,初步探究鹿茸提取物抗氧化作用及其机制。方法采用化学方法检测不同浓度鹿茸提取物清除DPPH、ABTS自由基的能力;以线虫为模型,通过氧化应激、热应激、紫外应激、寿命、运动能力、生殖能力等实验,初步探究其体内抗氧化作用及机制。结果鹿茸提取物具有清除DPPH、ABTS自由基的能力,且呈剂量依赖性;饲喂2.5 g·L^(-1)的鹿茸提取物使线虫在氧化应激条件下能显著延长生存时间,表明其具有抗氧化应激能力;进一步研究发现,加药组clk-1突变体线虫与对照组相比,在氧化应激下生存时间无明显差异,推测其抗氧化作用机制可能依赖于线粒体信号通路;鹿茸提取物还能增强抗热应激和抗紫外应激的能力,提高线虫运动和生殖能力,表明其具有抗衰老的作用。结论鹿茸提取物在体外、体内均具有抗氧化能力,减少氧化应激造成的损伤,进而延长寿命,起到延缓衰老的作用。 Objective To study the antioxidant activities and mechanisms of velvet antler extracts with Caenorhabditis elegans(C.elegans)as model organisms.Methods The abilities of scavenging DPPH and ABTS in different concentrations of velvet antler extracts were tested by chemical method;using C.elegans as the models,the antioxidant effects and mechanisms of velvet antler extracts in vivo were preliminarily studied by experiments of oxidative stress,heat stress,ultraviolet stress,life span,athletic ability and reproductive ability.Results Velvet antler extracts had the abilities of scavenging DPPH and ABTS,and their antioxidant effects in vitro were dose-dependent.Feeding 2.5 g·L^(-1)velvet antler extracts could significantly prolong the survival time of C.elegans under oxidative stress,which indicated the abilities of antioxidant stress of velvet antler extracts.Compared with the control groups,clk-1 mutant C.elegans in the treatment groups had no significant differences in the survival time under oxidative stress,suggesting that the antioxidant mechanisms might depend on mitochondrial signal pathways.Velvet antler extracts could also enhance the abilities to resist heat stress and ultraviolet stress,and improve the movements and reproductions abilities of C.elegans',which showed that they had the anti-aging effects.Conclusion Velvet antler extracts have the abilities of anti-oxidation in vitro and in vivo,reduce the damages caused by oxidative stress,prolong the life spans,and play the roles of anti-aging.
