微小RNA-335、癌胚抗原相关细胞黏附分子5在胃癌组织中的表达及与预后的关系

Expression of miR-335 and CEACAM5 in gastric cancer and its relationship with prognosis

目的 检测胃癌组织中微小RNA-335(miR-335)、癌胚抗原相关细胞黏附分子5(CEACAM5)表达情况,探讨与预后的关系。方法 取本院80例胃癌患者胃癌组织、癌旁组织分别为胃癌组、对照组,测定miR-335、CEACAM5mRNA表达水平和CEACAM5蛋白表达情况;分析miR-335、CEACAM5 mRNA相关性,miR-335、CEACAM5蛋白与临床病理特征关系,患者3年生存情况,影响预后的因素。结果 与对照组比较,胃癌组miR-335水平降低,CEACAM5mRNA水平及其蛋白阳性率升高(P<0. 05)。miR-335、CEACAM5 mRNA表达呈负相关(P<0. 05)。miR-335低表达者、CEACAM5蛋白阳性者Ⅲ期~Ⅳ期、低分化、发生淋巴结转移比例分别高于miR-335高表达者、CEACAM5蛋白阴性者(P<0. 05)。miR-335低表达者、CEACAM5蛋白阳性者3年累积存活率分别低于miR-335高表达者、CEACAM5蛋白阴性者(P<0. 05)。发生淋巴结转移、miR-335低表达、CEACAM5蛋白阳性是影响预后的危险因素(P<0. 05)。结论 miR-335低表达、CEACAM5蛋白阳性表达可能与胃癌发生发展有关,可一定程度评估预后。

Objective To detect the expression of microRNA-335(miR-335)and carcinoembryonic antigen-related cell adhesion molecule 5(CEACAM5)in patients with gastric cancer,and to explore the relationship between the expression of miR-335 and CEACAM5 and the prognosis of patients.Methods Gastric cancer tissues and adjacent tissues of 80 patients with gastric cancer were selected as gastric cancer group and control group.The levels of miR-335,CEACAM5 mRNA and CEACAM5 protein were determined;the correlation of miR-335 and CEACAM5 mRNA,the relationship between miR-335 and CEACAM5 protein and clinicopathological features,3-year survival and prognostic factors were analyzed.Results Compared with the control group,the level of miR-335 in gastric cancer group was lower,and the level of CEACAM5 mRNA and positive rate of CEACAM5 protein were higher(P<0.05).There was a negative correlation between miR-335 and CEACAM5 mRNA expression(P<0.05).Patients with low miR-335 expression,CEACAM5 positive,Ⅲ-Ⅳ stage,low differentiation and lymph node metastasis rate were higher than those with high miR-335 expression and CEACAM5 negative(P<0.05).The 3-year cumulative survival rates of low miR-335 expression and CEACAM5 protein positive group were lower than those of high miR-335 expression and CEACAM5 protein negative group respectively(P<0.05).Lymph node metastasis,low expression of miR-335 and positive expression of CEACAM5 protein were risk factors of prognosis in patients with gastric cancer(P<0.05).Conclusion The low expression of miR-335 and the positive expression of CEACAM5 protein may be related to the occurrence and development of gastric cancer,and they can evaluate the prognosis of patients to a certain extent.
