摘要
目的探究泛素水解酶22(USP-22)对舌鳞癌细胞侵袭与迁移的影响及其调控机制。方法收集2014年2月—2019年7月入住青岛大学附属医院口腔颌面外科的舌鳞癌患者21例,术中收集患者的肿瘤组织和癌旁组织,利用免疫组织化学染色检测两种组织中USP-22和细胞外信号调控蛋白激酶5(ERK5)蛋白的表达。构建USP-22基因siRNA沉默质粒,根据是否转染质粒,将细胞分为TCA8113细胞对照组(A组)、TCA8113细胞未转染组(B组)、TCA8113细胞转染组(C组)及Tb细胞对照组(D组)、Tb细胞未转染组(E组)、Tb细胞转染组(F组)。采用划痕实验和Transwell实验检测各组细胞的侵袭与迁移能力。结果高分化组和中低分化组舌鳞癌组织中USP-22蛋白和ERK5蛋白的表达量与对照组比较具有显著性差异(F=377.72、211.29,q=22.72~34.13,P<0.05)。A~C组、D~F组细胞迁移距离和侵袭能力比较均具有显著性差异(F=12.21~43.22,P<0.05),其中C组细胞迁移距离和侵袭能力明显低于A、B组(q=5.41~11.83,P<0.05),F组细胞迁移距离和侵袭能力明显低于D、E组(q=7.24~7.84,P<0.05)。结论USP-22对舌鳞癌细胞的侵袭和迁移能力具有一定的影响。
Objective To investigate the effect of ubiquitin-specific peptidase 22(USP-22)on the invasion and migration of tongue squamous carcinoma cells and its regulatory mechanism.Methods A total of 21 patients with tongue squamous carcinoma who were admitted to Department of Oral and Maxillofacial Surgery,The Affiliated Hospital of Qingdao University,from February 2014 to July 2019 were enrolled.Tumor tissue and adjacent tissue were collected from these patients,and immunohistochemical staining was used to measure the expressions of USP-22 and extracellular signal-regulated kinase 5(ERK5)in tumor tissue and adjacent tissue.The siRNA silencing plasmid of the USP-22 gene was constructed,and according to whether the cells were transfected with such plasmid,the cells were divided into TCA8113 cell control group(group A),TCA8113 cell non-transfection group(group B),TCA8113 cell transfection group(group C),Tb cell control group(group D),Tb cell non-transfection group(group E),and Tb cell transfection group(group F).Wound healing assay and Transwell assay were used to evaluate the migration and invasion abilities of cells in each group.Results The protein expression levels of USP-22 and ERK5 in tongue squamous carcinoma tissue in the well-differentiated group and the moderately-to-poorly differentiated group were significantly different from those in the control group(F=377.72,211.29,q=22.72-34.13,P<0.05).There were significant differences in cell migration distance and invasion ability between groups A-C and groups D-F(F=12.21-43.22,P<0.05);group C had a significantly shorter cell migration distance and a significantly weaker invasion ability than groups A and B(q=5.41-11.83,P<0.05),and group F had a significantly shorter cell migration distance and a significantly weaker invasion ability than groups D and E(q=7.24-7.84,P<0.05).Conclusion USP-22 has a certain effect on the invasion and migration abilities of tongue squamous carcinoma cells.
