摘要
目的利用高效液相色谱法与色差法对紫锥菊药材进行质量评价研究,探讨药材颜色与成分含量之间的相关性,为紫锥菊的质量评价提供一种简便快速的新方法。方法采用NH300分光测色仪对10批紫锥菊药材进行色差值的测量,运用单因素分析法对测量的色差值进行差异性分析,运用HPLC-UV法测定不同批次的紫锥菊药材中菊苣酸含量,结合聚类分析法对不同批次紫锥菊药材进行质量评价。结果采用单因素方差分析的方法比较ΔE*值(总色差值)可以看出10批紫锥菊在色差方面有非常显著性差异,运用聚类分析法发现随着△α*值的改变,菊苣酸的含量也随之改变,进而对不同批次紫锥菊的质量进行评价。属于绿-黄色域的药材中菊苣酸含量在10批药材中含量最高(平均含量为1.01%),属于淡绿-黄色域的次之(平均含量为0.82%),属于红-黄色域的含量最低(平均含量为0.52%)。通过分类发现,在6至10月采收的药材质量明显优于11月采收的药材。结论HPLC-UV法与色差法操作简便,容易实现,并且结合聚类分析法可有效地评价出不同批次紫锥菊的质量。两种方法的结合为未来紫锥菊的鉴定、质量评价提供新的研究思路。
Objective To evaluate the quality of Echinacea purpurea by high performance liquid chromatography(HPLC)and color difference method,and to explore the correlation between the color and the content of components,so as to provide a simple and rapid new method for the quality evaluation of Echinacea purpurea.Methods Ten batches of Echinacea purpurea were measured by NH300 spectrophotometer.The difference of color difference was analyzed by single factor analysis.The content of cichoric acid in different batches of Echinacea purpurea was determined by HPLC-UV.The quality of different batches of Echinacea purpurea was evaluated by cluster analysis.Results The difference ofΔE;value(total color difference)between 10 batches of Echinacea purpurea was significant by one-way ANOVA.Cluster analysis showed that with the change ofΔα;,the content of cichoric acid also changed,and then the quality of different batches of Echinacea purpurea was evaluated.The content of cichoric acid in 10 batches of herbs belonging to green-yellow region was the highest(average content was 1.0121%),followed by light green-yellow region(average content was 0.8199%),and the content of cichoric acid in red-yellow region was the lowest(average content was 0.5234%).Through classification,it was found that the quality of medicinal materials collected from June to October was significantly better than that collected in November.Conclusion HPLC-UV method and color difference method are simple and easy to implement,and combined with cluster analysis method can effectively evaluate the quality of different batches of Echinacea purpurea.The combination of the two methods provides a new research idea for the identification and quality evaluation of Echinacea purpurea in the future.
