引经药配伍对血府逐瘀汤预防非酒精性脂肪肝病作用的影响

Influence of Compatibility of Guiding-herb on the Effect of Xuefu Zhuyu Decoction in Preventing Nonalcoholic Fatty Liver Disease

目的观察血府逐瘀汤配伍引经药桔梗、牛膝对非酒精性脂肪肝(NAFLD)的病理形态学影响。方法将36只C57BL/6J ApoE^(-/-)小鼠随机分出6只为空白组,余下小鼠采用腹腔注射亚临床剂量LPS(4ng·kg^(-1))联合高脂饲料喂养建立NAFLD模型,并按体重排序法进行如下分组:模型组、血府逐瘀汤组、血府逐瘀汤减牛膝组、血府逐瘀汤减桔梗组、血府逐瘀汤减牛膝-桔梗组,造模期间分别给予血府逐瘀汤、血府逐瘀汤减牛膝方、血府逐瘀汤减桔梗方、血府逐瘀汤减牛膝-桔梗方灌胃干预28 d。肝组织HE染色和油红O染色观察小鼠肝组织病理形态学改变。结果与模型组相比,血府逐瘀汤及其拆方组可降低小鼠肝脏肿胀程度;与模型组相比,血府逐瘀汤及其拆方组肝组织脂肪变性与小叶炎症评分略有降低(P>0.05);与模型组和血府逐瘀汤组比较,血府逐瘀汤减牛膝-桔梗组的肝脏组织脂滴百分比显著升高(P<0.01)。结论血府逐瘀汤能改善NAFLD模型小鼠中性粒细胞浸润,从病理上改善肝组织的损伤,其中引经药牛膝、桔梗在预防NAFLD中起到明显增效作用。

Objective Observation of the pathological morphology of Xuefu Zhuyu Decoction combined with guiding-herb on non-alcoholic fatty liver(NAFLD).Methods 36 male C57 BL/6 J ApoE^(-/-)mice were randomly divided into 6 mice as the blank group,the remaining mice were fed by intraperitoneal injection of subclinical dose of LPS(4 ng·kg^(-1))combined with high-fat diet to establish NAFLD model,and grouped according to the weight ranking method as follows:model group,Xuefu Zhuyu Decoction group,Xuefu Zhuyu Decoction minus Achyranthes bidentata group,Xuefu Zhuyu Decoction minus Platycodon grandiflorum group,Xuefu Zhuyu Decoction minus Achyranthes bidentata,Platycodon grandiflorum group.During the modeling period,each prevention group was given the Xuefu Zhuyu prescription,Xuefu Zhuyu Decoction minus Achyranthes bidentata prescription,Xuefu Zhuyu Decoction minus Platycodon grandiflorum prescription,Xuefu Zhuyu Decoction minus Achyranthes bidentata-phatycodon prescription gavage intervention for 28 days.Compared with the appearance of the mouse liver,hematoxylin-eosin staining(HE staining)of the left outer leaf of the liver and the staining with oil red O in liver were used to observe the pathological changes of the liver.Results Compared with the model group,Xuefu Zhuyu Decoction and its decomposed formula group can reduce the degree of liver swelling in mice;Compared with the model group,the scores of Xuefu Zhuyu Decoction and its decomposed formulas group for liver tissue steatosis and lobular inflammation were slightly reduced(P>0.05);Compared with the model group and the Xuefu Zhuyu Decoction group,Xuefu Zhuyu Decoction reduced the liver tissue of Achyranthes bidentata.The percentage of lipid droplets increased significantly(P<0.01).Conclusion Xuefu Zhuyu Decoction can improve the infiltration of neutrophils in NAFLD model mice and pathologically improve liver tissue damage.Among them,the meridian drugs Achyranthes bidentata and Platycodon grandiflorum play a significant synergistic effect in preventing NAFLD.
