摘要
目的建立甘姜苓术汤多指标性成分含量测定方法,采用正交实验设计优选最佳煎煮工艺,还原古代经典名方甘姜苓术汤物质基准制备方法,以标准化工艺控制甘姜苓术汤质量稳定。方法在与古籍记载一致的前提下,结合现代科研技术,实验以甘草苷、甘草酸、6-姜辣素为指标性成分,建立甘姜苓术汤HPLC含量测定方法。选取浸泡时间、煎煮火力、砂锅是否加盖三个因素,以干浸膏得率和指标性成分含量为评价指标,进行L_(9)(3^(4))正交优选,制定合理工艺参数,得到最佳甘姜苓术汤煎煮工艺并进行工艺验证。结果甘姜苓术汤的最佳煎煮工艺为加水1000 ml,浸泡60 min,以武火(2200W)煮沸后,半开盖,调节火力至800W,保持微沸并煮至600 ml趁热过滤。干浸膏得率为19.51%,甘草苷、甘草酸、6-姜辣素含量分别为1.61%、2.12%、0.036%,工艺验证后RSD均小于3%,表明此煎煮工艺稳定可行。结论该工艺为甘姜苓术汤最佳煎煮工艺,为甘姜苓术汤物质基准研究提供科学工艺制备方法。
Objective To establish a determination for multiple index components of Ganjianglingzhu Decoction.Using orthogonal experiment design to optimize the best decocting process to restore preparation method of substance bechmark of Ganjianglingzhu Decoction,and stablely control quality with standardized processes.Methods On the premise that it is consistent with records in ancient books,combined with modern scientific research technology,experiment uses Liquiritin,Glycyrrhizic acid and 6-Gingerol as the index components to establish a HPLC method for the determination of Ganjianglingzhu Decoction.Choosing the three factors like soaking time,decoction firepower,and whether the casserole is covered or not,and taking the yield of dry extract and content of index components as evaluation indicators to carry out L_(9)(3^(4))orthogonal optimization and formulate reasonable process parameters for obtaining the best decocting process and verifying the process.Results The best decocting process of Ganjianglingzhu Decoction is to add 1000 ml of water,soak for 60 minutes,boil(2200 W),half open the lid,slightly boil(800 W)and boil it until 600 ml,filter it.The yield of dry extract of Ganjianglingzhu Decoction is 19.51%,and the contents of Liquiritin,Glycyrrhizic acid and 6-Gingerol are 1.61%,2.12%,and 0.036%respectively.After the process verification,the RSD is less than 3%,indicating that the decocting process is stable and feasible.Conclusion The best decocting process of Ganjianglingzhu Decoction provides a scientific preparation method for research on the substance benchmark of Ganjianglingzhu Decoction.
