三元两相溶剂系统提取中药虎杖中白藜芦醇的工艺研究

摘要 目的:优选三元两相溶剂提取中药虎杖中白藜芦醇的工艺条件。方法:采用正交试验和单因素试验,通过薄层扫描外标法进行含量测定,以白藜芦醇提取率和纯度为考察指标,优选提取工艺条件。结果:最佳提取工艺为:丙酮︰水(7︰3)溶液按照1︰10料液比超声提取两次,每次15 min,调粗提液pH至8.5,加入3/5体积二氯甲烷,充分振摇,35℃下静置分层40 min。工艺验证结果表明经一步萃取后白藜芦醇纯度可达46%,提取率为4.1‰,RSD分别为0.97%和2.19%。结论:优选工艺简单高效,稳定可行。 Objective:To optimizing the technology condition of ternary two-phase liquid–liquid extraction for resveratrol from Polygonum Cuspidatum.Method:The content of resveratrol was determined by TLC,and the extraction yield and purity of resveratrol were regarded as the index.Orthogonal test and a single factor test were used to optimize the extraction process of resveratrol in Polygonum cuspidatum.Results:The optimum extraction condition is that the ratio of material to liquid(acetone/water=7/3)is 1︰10,the extraction is twice and 15 minutes per time by ultrasonic,the pH of the solution is adjusted to 8.5 and the volume of dichloromethane is 3/5 of acetone/water,the phase separation temperature is at 35℃for 40 min.Results shows that only for one extraction step the purity of resveratrol is 46%,and the extraction yield is 4.1‰,the RSD were 0.97%and 2.19%respectively.Conclusion:The optimum technology condition is uncomplicated,efficient,stable and feasible.
