摘要
目的:探讨红豆杉提取物维康醇抑制膀胱癌T24细胞增殖的机制。方法:用维康醇处理膀胱癌T24细胞,MTS法检测维康醇对细胞增殖的影响;流式细胞术检测维康醇对细胞周期的影响;实时荧光定量PCR和westernblot检测Cyclin D1、CDK4、p21 mRNA和蛋白表达水平的影响。结果:维康醇呈浓度和时间依赖性抑制T24细胞的增殖;维康醇诱导细胞周期阻滞于G1期,并下调Cyclin D1、CDK4表达,上调p21表达。结论:维康醇能够抑制膀胱癌T24细胞的增殖,可能与下调Cyclin D1和CDK4表达,上调p21表达,导致细胞周期于G1期阻滞有关。
Objective To investigate the mechanism of the inhibitory effect of Alteronol on the proliferation of bladder cancer T24 cells.Methods Bladder cancer T24 cells were treated with Alteronol.MTS assay was performed to test the effect of Alteronol on cell proliferation.Flow cytometry was used to detect the effect of Alteronol on cell cycle,and real-time quantitative PCR and western blot were performed to detect the expression of cyclin D1,CDK4 and p21.Results Alteronol inhibited the proliferation of T24 cells in a concentration and time-dependent manner.Alteronol induced cell arrest in G1 phase,down-regulated the expression of cyclin D1 and CDK4,and up-regulated the expression of p21.Conclusion Alteronol can inhibit the proliferation of bladder cancer T24 cells,which may be related to the down-regulation of the expression of Cyclin D1 and CDK4 and up-regulation of p21 expression,leading to cell cycle arrest in G1 phase.
