苍耳子散对变应性鼻炎小鼠炎症及AQP5蛋白表达的影响 被引量:7

摘要 目的:研究苍耳子散对变应性鼻炎(AR)小鼠鼻腔炎症及水通道蛋白5(AQP5)表达的影响。方法:将60只小鼠随机分成正常组、模型组、氯雷他定组、苍耳散低剂量组、苍耳散中剂量组和苍耳散高剂量组,每组10只,除正常组外,其他各组小鼠制备AR模型,氯雷他定组灌胃氯雷他定药液,苍耳子散低、中、高剂量组分别灌胃相应剂量苍耳子散药液,连续给药14 d,每天一次。观察各组小鼠体质量、挠鼻次数、打喷嚏次数、脏器指数、采用ELISA法检测小鼠血清中IgE、IL-5、IL-4、IFN-γ、HIS和IL-10含量,利用Western Blot法检测鼻黏膜组织汇总AQP5蛋白表达水平。结果:与正常组相比,模型组小鼠终末体质量、IFN-γ、IFN-γ/IL-4和AQP5蛋白表达水平显著性降低(P<0.05,P<0.01),小鼠挠鼻次数,打喷嚏次数,脏器指数,IgE、IL-5、IL-4、HIS和IL-10含量均显著性增加(P<0.05,P<0.01)。与模型组相比,苍耳子散中剂量组小鼠挠鼻次数,打喷嚏次数,脾脏指数,血清中IL-4、IgE、HIS、IL-5和IL-10含量显著性降低(P<0.05,P<0.01),苍耳子散高剂量组小鼠挠鼻次数,打喷嚏次数,脾脏指数,血清中IL-4、IgE、HIS、IL-5和IL-10含量显著性降低(P<0.05,P<0.01),而小鼠终末体质量,IFN-γ、IFN-γ/IL-4和AQP5蛋白表达水平显著性增加(P<0.05,P<0.01)。结论:苍耳子散可显著缓解AR小鼠临床症状,降低血清炎性因子含量,其机制可能与调节Th细胞分泌细胞因子及AQP5蛋白表达有关。 Objective To study the effect of Xanthium powder on nasal inflammation and AQP5 expression in allergic rhinitis(AR)mice.Methods 60 mice were randomly divided into normal group,model group,loratadine group,Xanthium powder low-dose group,Xanthium powder medium dose group and Xanthium powder high-dose group,10 in each group,In addition to the normal group,other groups of mice made AR model,the loratadine group was given loratadine solution by gavage,while the low,medium,and high-dose Xanthium powder groups were intragastrically administered with corresponding doses of Xanthium powder,once a day for 14 consecutive days.The body weight,nasal scratching times,sneezing times,organ index,IgE,IL-5,IL-4,IFN-γ,HIS and IL-10 in serum were detected by ELISA,and AQP5 protein expression in nasal mucosa was detected by Western blot.Result Compared with the normal group,the weight of terminal body,the expression of IFN-γ,IFN-γ/IL-4 and AQP5 protein in the model group were significantly decreased(P<0.05,P<0.01),and the number of nose scratching,sneezing frequency,organ index,IgE,IL-5,IL-4,HIS and IL-10 were significantly increased(P<0.05,P<0.01).Compared with the model group,the number of scratching nose,sneezing frequency,spleen index,contents of IL-4,IgE,HIS,IL-5 and IL-10 in serum were significantly decreased in the middle dose group(P<0.05,P<0.01),while the nasal scratching times,sneezing times,spleen index,IL-4,IgE,HIS,IL-5 and IL-10 contents in serum were significantly decreased in the high-dose Xanthium powder group(P<0.05,P<0.01),However,the weight of mouse terminal body,the expression of IFN-γ,IFN-γ/IL-4 and AQP5 protein were significantly increased(P<0.05,P<0.01).Conclusion Cangerzi powder can significantly alleviate the clinical symptoms of AR mice and reduce the content of serum inflammatory factors,which may be related to the expression of AQP5 protein.
