ARTP-UV复合诱变选育高产木聚糖酶菌株

摘要 以黑曲霉G-1为出发菌株,通过常温常压等离子体(ARTP)和紫外线(UV)复合诱变技术,筛选到1株酶活提高明显的菌株GAU10-18。经过多次摇瓶复筛验证,其遗传性质稳定,酶活提高明显,摇瓶发酵酶活达到25000 U/mL,较出发菌株提高95%左右。另外,该菌株所产的酸性木聚糖酶具有良好的耐热、耐酸性,在较低温度仍具有很高的酶活性。 The original strain of Aspergillus niger G-1 was modified in genetic characteristics with compound mutagenesis by ARTP and UV.A high glucoamylase-yielding strain GAU10-18 was screened from the mutants by slab screening and shake flask rescreening,which has high enzyme activity and good genetic stability.The enzyme activity of the shake flask-fermentation reached 25000u/ml,which was 95%higher than that of the original strain.In addition,the acid xylanase produced by this strain has good heat resistance,acid and alkali resistance,and high enzymatic activity at lower temperature.
