氟尿嘧啶热化疗对结肠癌细胞的细胞毒作用 被引量:1

摘要 目的:研究5-氟尿嘧啶(5-Fu)热化疗对结肠癌HT-29细胞的细胞毒作用。方法:将HT-29细胞分为对照组、5-Fu组、5-Fu热化疗组,对照组不作任何处理,5-Fu组加入未加热5-Fu,5-Fu热化疗组加入43℃5-Fu,采用CCK8法检测3组HT-29细胞增殖率,采用流式细胞术检测3组HT-29细胞凋亡率,通过Transwell小室实验检测3组HT-29细胞侵袭能力。结果:5-Fu组HT-29细胞增殖率明显低于对照组(P<0.05);5-Fu热化疗组HT-29细胞增殖率明显低于5-Fu组(P<0.05);5-Fu组HT-29细胞凋亡率明显高于对照组(P<0.05);5-Fu热化疗组HT-29细胞凋亡率明显高于5-Fu组(P<0.05);5-Fu组穿过小室膜的HT-29细胞数明显少于对照组(P<0.05);5-Fu热化疗组穿过小室膜的HT-29细胞数明显少于5-Fu组(P<0.05)。结论:5-Fu热化疗对结肠癌HT-29细胞具有明显毒作用,5-Fu热化疗能够抑制结肠癌细胞增殖及侵袭,诱导结肠癌细胞凋亡,这可为临床治疗结肠癌提供一定参考。 Objective:To investigate the cytotoxicity of 5-fluorouracil(5-Fu)thermo-chemotherapy on colon cancer HT-29 cells.Methods:HT-29 cells were divided into three groups for different treatments,namely,control group,5-FU group and 5-FU thermochemotherapy group.After treatment,the proliferation rate,apoptotic rate and invasive ability of HT-29 cells in three groups were detected by CCK8 method,flow cytometry and transwell chamber.Results:The proliferation rate of HT-29 cells was the highest in control group,followed by 5-FU group and 5-FU thermochemotherapy group,with statistic difference(P<0.05).The apoptosis rate of HT-29 cells was the lowest in control group,followed by 5-FU group and 5-FU thermochemotherapy group,with statistic difference(P<0.05).The number of HT-29 cells passing through the ventricular membrane was the highest in control group,followed by 5-FU group and 5-FU thermochemotherapy group,with statistic difference(P<0.05).Conclusion:5-fluorouracil thermochemotherapy has obvious toxic effect on colon cancer HT-29 cells,which can inhibit the proliferation and invasion of colon cancer cells and induce apoptosis of colon cancer cells.
