芪斛楂颗粒对脾虚模型大鼠胃肠功能及免疫系统的影响 被引量:4

摘要 目的探讨芪斛楂颗粒对脾虚模型大鼠胃肠功能及免疫系统的影响。方法取雄性SD大鼠30只,采用饮食不节、过度疲劳和苦寒破气相结合的复合因素法建立脾虚大鼠模型,于造模成功后将脾虚大鼠随机分为模型组、阳性对照组(补中益气颗粒630 mg/kg)和芪斛楂颗粒组(芪斛楂颗粒200 mg/kg),每组10只,另取10只同批次未造模大鼠作为对照组(生理盐水5 mL/kg);末次给药后观察大鼠外观行为变化并评分,随后取血及取各组大鼠的胸腺及脾脏,计算胸腺指数及脾指数,采用酶联免疫吸附法(ELISA)检测各组大鼠血清中胃泌素(GAS)、胃动素(MTL)、生长抑素(SS)、肿瘤坏死因子(TNF-α)和白细胞介素-1β(IL-1β)的含量。结果与对照组比较,模型组大鼠外观行为学评分明显升高(P<0.01),体质量、胸腺指数、脾指数、血清GAS、MTL含量均明显降低(P<0.01),血清SS、TNF-α、IL-1β含量明显上升(P<0.01);与模型组比较,阳性对照组和芪斛楂颗粒组大鼠外观行为学评分值显著下降(P<0.01),体质量、胸腺指数、脾指数、血清中的GAS、MTL含量均明显升高(P<0.01),血清SS、TNF-α、IL-1β含量明显降低(P<0.01)。结论芪斛楂颗粒对脾虚模型大鼠具有改善胃肠功能及调节免疫系统功能的作用,其机制可能与提高血液中GAS、MTL含量,降低SS、TNF-α、IL-1β含量有关。 Objective To investigate the effect of qihuzha granules on gastrointestinal function and immune system of spleen-deficiency rats.Methods The compound factor method of bitter cold diarrhea,irregular diet and overfatigue was used to build the spleen-deficiency model of male SD rats.After the model was established,model rats were randomly divided into the model group,the positive control group(buzhongyiqi granules,630 mg/kg),and qihuzha granules group(qihuzha granules,200 mg/kg),with 10 rats in each group.Other 10 unmodeled rats of the same batch served as the control group(normal saline,5 mL/kg).All the rates in each group were given corresponding drugs once a day for consecutive 10 days.During the experiment,the appearance and behavior changes of the rats were observed.At the end of the experiment,the index of thymus and spleen were calculated,the levels of serum gastrin(GAS),motilin(MTL),somatostatin(SS),tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)of rats were detected by the enzyme-linked immunosorbent assay(ELISA).Results Compared with the control group,the model group’s appearance behavior score significantly increased(P<0.01),while their body weight,the index of thymus and spleen,levels of serum GAS,MTL(P<0.01)significantly decreased,and the level of serum SS,TNF-α,IL-1βobviously increased(P<0.01).Compared with the model group,the appearance and behavior scores of qihuzha granules group were significantly reduced(P<0.01),while the body weight,the index of thymus and spleen,and the levels of serum GAS,MTL(P<0.01)and the level of serum SS,TNF-α,IL-1βsignificantly decreased(P<0.01).Conclusion Qihuzha granules can improve the gastrointestinal function and regulate the immune system in spleen-deficiency model rats.It may be related to the increase of the content of GAS,MTL,and the index of thymus and spleen,and to the reduction of the content of SS,TNF-α,and IL-1β.
