春雷霉素在玉米、烟草中的吸收与转运 被引量:1

Uptake and Transportation of Kasugamycin in Corn and Tobacco

摘要 [目的]探索春雷霉素(KSM)在植物体的吸收与转运特性。[方法]通过浸叶法处理烟草和玉米幼苗中部叶片,采用高效液相色谱串联质谱仪检测KSM在不同处理下烟草和玉米幼苗顶端叶、茎及根部的含量;通过4-氯-7-硝基-2,1,3-苯并氧杂二唑(NBD-Cl)与KSM偶合,生成荧光偶合物KSM-NBD示踪KSM在植物体的传导途径。[结果]温度、pH值、能量抑制剂和糖转运蛋白竞争底物均能影响KSM的吸收;荧光显像表明,烟草和玉米的顶端叶、茎及根中KSM-NBD主要分布于韧皮部和木质部。[结论]KSM受载体蛋白协助进入植物组织,且通过木质部和韧皮部双向传导,为后期开发双向输导型农药,提供思路和理论支撑;为指导田间科学施用KSM防治作物病害提供理论基础。 [Aims]This study aims to explore the absorption and transport characteristics of kasugamycin(KSM)in plants.[Methods]The middle leaves of tobacco and maize were treated by the leaf soaking method,and high performance liquid chromatography tandem mass spectrometer was used to detect the contents in leaves,stems and roots of the tobacco and corn seedlings under different treatments by KSM;through 4-chloro-7-nitro-2,1,3-benzoxoxadiazole(NBD-Cl)to form the fluorescent conjugate KSM-NBD trace the conduction pathway of KSM in plants.[Results]Temperature,pH,energy inhibitors and sugar transporter competing substrates can all affect the absorption of KSM.The fluorescence imaging showed that KSM-NBD in the apical leaves,stems and roots of tobacco and corn are mainly distributed in phloem and xylem.[Conclusions]KSM is assisted by carbohydrate carrier protein to enter plant tissues,and conducts bidirectionally through the xylem and phloem,which provides ideas and theoretical support for further development of bidirectional pesticides;and provides a theoretical basis for scientific application of KSM in the field to prevent and control crop diseases.
