节杆菌S-4的鉴定及其生物脱硫特性研究 被引量:2

摘要 水体中的硫化物以及天然气和沼气中含有的硫化氢不仅污染环境,而且危害人体健康。为了探索绿色清洁的生物除硫方法,一株具有硫化物氧化能力的细菌S-4从硫铁矿石表面的土样中分离获得。该研究通过观察菌株S-4的菌落形态特征以及比对16S rRNA序列,得知菌株S-4属于节杆菌属(Arthrobacter sp.)。单因素试验和Box–Behnken响应面优化试验对其生物脱硫特性进行研究,结果表明菌株S-4对培养温度和初始pH值较敏感,在初始pH 7.4、30℃、培养66 h的条件下,菌株S-4生长速率快,S2-氧化率高,达到S2-最大氧化率93.84%,在生物脱硫等领域具有一定的应用价值。 Sulphide in water and hydrogen sulfide contained in natural gas and biogas causes not only environment pollution but also the risks to human health.In order to explore the approaches to green and clean biological sulfur removing,a strain S-4 with sulfide oxidation ability was isolated from the soil samples taken from the surface of pyrite ore,and the biological desulfurization characteristics were investigated.By virtue of observation of the morphological characteristics of strain S-4 and in comparison with the results of 16 S rRNA sequence,the strain S-4 was identified as Arthrobacter sp.The single factor test and Box-Behnken response surface optimization test showed that strain S-4 was sensitive to culture temperature and initial pH value.Under the condition such as initial pH 7.4,culture temperature 30℃and time 66 h,the growth rate of train S-4 was fast,and S2-oxidation rate high,the maximum S^(2-)oxidation rate could be as high as 93.84%.
