摘要
目的:通过生物信息学方法分析睾素基因家族(sparc/osteonectin, cwcv and kazal-like domains proteoglycan, SPOCK)在卵巢癌组织中的表达及临床意义。方法:利用GEPIA和UALCAN数据库比较及验证SPOCK在正常卵巢组织及卵巢癌组织中的表达差异;利用GEPIA数据库评价SPOCK2表达对卵巢癌患者预后的影响;利用GeneMANIA网站预测SPOCK2的蛋白质相互作用(protein-protein interaction, PPI)网络;利用DAVID对SPOCK2及其关键的互作基因进行基因富集(gene ontology, GO)分析和通路富集(kyotoencyclopedia of genes and genomes, KEGG)分析。结果:在GEPIA数据库中,与正常卵巢组织相比,SPOCK2在卵巢癌组织中显著上调,而SPOCK、SPOCK3无显著差异表达。通过CPTAC数据库进一步确认SPOCK2蛋白水平在卵巢癌组织中高表达;此外,SPOCK2高表达患者的总生存率(overall survival, OS)均明显低于SPOCK2低表达患者。蛋白质互作网络分析揭示了包括SPOCK3、PLXNB3在内的20个关键互作分子;GO分析和KEGG分析揭示了SPOCK2及其关键互作分子的潜在分子机制。结论:SPOCK2在卵巢癌组织中高表达,且与卵巢癌的发生、发展及预后密切相关,可以作为卵巢癌诊断、预后预测的标志物之一。
Objective:To analyze the expression and clinical significance of sparc/osteonectin,cwcv and kazal-like domains proteoglycan(SPOCK)family in ovarian cancer tissues by bioinformatics methods.Methods:GEPIA and UALCAN databases were used to compare and verify the expression differences of SPOCK in normal ovarian and ovarian cancer tissues.The GEPIA database was used to evaluate the prognostic value of SPOCK2 expression in ovarian cancer patients.The GeneMANIA website was used to predict the protein-protein interaction(PPI)network of SPOCK2.DAVID was applied to performgene ontology(GO)and kyotoencyclopedia of genes and genomes(KEGG)analysis of SPOCK2 and its key interacted genes.Results:In GEPIA database,SPOCK2 was significantly upregulated in ovarian cancer tissues compared with normal ovarian tissues,while SPOCK1 and SPOCK3 were not significantly differentially expressed.The SPOCK2 protein level was further confirmed to be highly expressed in ovarian cancer by the CPTAC database.In addition,the overall survival(OS)of patients with high SPOCK2 expression was significantly lower than those with low SPOCK2 expression.PPI network analysis revealed 20 key interaction molecules which included SPOCK3 and PLXNB3.GO and KEGG analysis revealed the potential molecular mechanism of SPOCK2 and its key interaction molecules.Conclusion:SPOCK2 is highly expressed in ovarian cancer tissues and is closely related to the oncogenesis,development and prognosis of ovarian cancer,which could be used as a biomarker for the diagnosis and prognostic evaluation of ovarian cancer.
