摘要
目的:探究儿童淋巴瘤患者化疗前后血液中长链非编码RNA GAS5(lncRNA GAS5)水平变化的临床意义。方法:收集淋巴瘤患儿(81例)及健康儿童(52例)的血液样本,采用实时荧光定量PCR(qRT-PCR)检测两组样本血液中的lncRNA GAS5含量。分析不同临床分期儿童淋巴瘤化疗有效和无效患儿血液中lncRNA GAS5表达及凝血功能在化疗前后的变化差异。淋巴瘤患儿血液中lncRNA GAS5水平与凝血指标的相关性利用皮尔森(Pearson)相关性分析。结果:患儿组lncRNA GAS5水平与凝血指标均显著高于对照组(P<0.01);淋巴瘤Ⅰ-Ⅱ期(n=45)与Ⅲ-Ⅳ期(n=36)患儿的lncRNA GAS5水平以及凝血功能间无统计学差异(P>0.05);化疗有效患儿(n=60)化疗后凝血指标及lncRNA GAS5水平与化疗前相比均显著降低(P<0.01);而化疗无效患儿组凝血指标lncRNA GAS5水平化疗前后差异无统计学意义(P>0.05)。lncRNA GAS5水平分别与PT(r^(2)=0.792)、APTT(r^(2)=0.776)和FIB含量(r^(2)=0.750)呈正相关(P<0.001)。结论:血液中lncRNA GAS5水平可能是反映淋巴瘤患儿化疗疗效和血栓发生风险的潜在标志物。
Objective:To explore the clinical significance of long noncoding RNA GAS5(lncRNA GAS5)in children with lymphoma.Methods:Blood samples of children with lymphoma(81 cases)and healthy children(52 cases)were collected,and qRT-PCR was used to detect the differences of lncRNA GAS5 content.The changes of lncRNA GAS5 expression and blood coagulation function before and after chemotherapy were analyzed,between effective and ineffective group in different clinical stages of childhood lymphoma.Pearson correlation coefficient method was used to analyze the correlation between blood lncRNA GAS5 level and coagulation index in children with lymphoma.Results:The lncRNA GAS5 level and coagulation index of the children group were significantly higher than those of the healthy group(P<0.01).There was no statistical difference between the children with lymphoma stageⅠ-Ⅱ(n=45)and stageⅢ-Ⅳ(n=36)(P>0.05).The lncRNA GAS5 levels and coagulation indexes of chemotherapy-effective children(n=60)after chemotherapy were significantly lower than those before chemotherapy(P<0.01).There was no statistically significant difference between before and after chemotherapy in children with chemotherapy-ineffective group(P>0.05).Moreover,lncRNA GAS5 levels were positively correlated with PT(r^(2)=0.77),APTT(r^(2)=0.75)and FIB content(r^(2)=0.79)(P<0.001).Conclusion:lncRNA GAS5 level in blood may be a potential marker reflecting the efficacy of chemotherapy and the risk of thrombosis in children with lymphoma.
