槲皮素对系统性红斑狼疮模型小鼠肾脏的保护作用及对其免疫功能的影响研究 被引量:9

摘要 目的研究槲皮素治疗对系统性红斑狼疮(SLE)模型小鼠肾脏的保护作用,并探讨槲皮素治疗对模型小鼠免疫功能的影响。方法将30只BALB/c雌性,体重16~22 g,6~8周龄小鼠按随机数字表法分为3组,即对照组、模型组和治疗组,每组10只。模型组和治疗组小鼠通过腹腔注射将值烷建立SLE小鼠模型,对照组小鼠给予腹腔注射0.9%氯化钠溶液;治疗组小鼠给予槲皮素灌胃治疗,对照组和模型组给予等量的0.9%氯化钠溶液。比较3组小鼠尿酸氮(BUN)、血肌酐(Scr)、24 h尿蛋白定量(24 h UP)以及肾脏肥大指数等肾功能指标,肾组织病理评分、免疫器官指数以及脾脏树突状细胞、辅助性T淋巴细胞亚群比例。结果治疗20周后,与对照组小鼠相比,模型组和治疗组小鼠BUN、Scr、24 h UP、肾脏肥大指数、肾小球评分、肾小管间质评分以及肾小管评分均显著升高,差异均有统计学意义(P<0.05);与模型组小鼠相比,治疗组小鼠BUN、Scr、24 h UP、肾脏肥大指数、肾小球评分、肾小管间质评分以及肾小管评分均显著降低,差异均有统计学意义(P<0.05)。治疗20周后,与对照组小鼠相比,模型组和治疗组小鼠脾脏重量、脾脏指数、胸腺重量、胸腺指数、脾脏树突状细胞、Th1淋巴细胞以及Th1/Th2细胞比例均显著降低,而脾脏Th2细胞比例显著升高,差异均有统计学意义(P<0.05);与模型组小鼠相比,治疗组小鼠脾脏重量、脾脏指数、胸腺重量、胸腺指数、脾脏树突状细胞、Th1淋巴细胞以及Th1/Th2细胞比例均显著升高,而脾脏Th2细胞比例显著下降,差异均有统计学意义(P<0.05)。结论槲皮素治疗可有效保护系统性红斑狼疮肾脏功能,改善其免疫功能。 Objective To study the protective effect of quercetin treatment on the kidneys of systemic lupus erythematosus(SLE)model mice,and to explore the effect of quercetin treatment on the immune function of the model mice.Methods Thirty BALB/c females,weighing 16~22 g,6~8 weeks old mice were divided into 3 groups by random number table method,namely control group,model group and treatment group,each group with 10 mice.Mice in the model group and the treatment group were given intraperitoneal injections of alkanes to establish SLE mouse models.The mice in the control group were given intraperitoneal injections of normal saline;the mice in the treatment group were given quercetin gavage,and the control group and the model group were given the same amount of saline.The blood urea nitrogen(BUN),serum creatinine(Scr),24-hour urine protein quantitative(24 h UP)and renal hypertrophy index and other renal function indexes,renal tissue pathological score,immune organ index and splenic dendritic cells,Proportion of helper T lymphocyte subsets of mice in 3 groups were compared.Results After 20 weeks of treatment,compared with mice in the control group,the BUN,Scr,24 h UP,renal hypertrophy index,glomerular score,tubular interstitial score and tubular score of the model group and the treatment group were significantly increased,and the differences were statistically significant(P<0.05),and the BUN,Scr,24 h UP,renal hypertrophy index,glomerular score,tubular interstitial score and tubular score of the treatment group were significantly decreased,and the differences were statistically significant(P<0.05).After 20 weeks of treatment,compared with the control group,the spleen weight,spleen index,thymus weight,thymus index,splenic dendritic cells,Th1 lymphocytes and Th1/Th2 cell ratios of the model group and the treatment group were significantly decreased,while the proportion of Th2 cells in the spleen were significantly increased,and the differences were statistically significant(P<0.05);compared with the model group,the spleen weight,spleen index,thymus weight,thymus index,splenic dendritic cells,Th1 lymphocytes and Th1/Th2 ratio of mice in the treatment group were significantly increased,while the ratio of Th2 cells in the spleen was significant decreased,the differences were statistically significant(P<0.05).Conclusion Quercetin treatment can effectively protect the kidney function of systemic lupus erythematosus and improve its immune function.
