摘要
目的探讨肉碱棕榈酰转移酶-2(CPT2)在卵巢上皮癌组织中的表达及临床意义,为研究卵巢癌的治疗方法提供新的靶点。方法收集2015年3月至2016年12月在西京医院接受治疗的20例卵巢上皮癌患者的新鲜组织标本,运用qRT-PCR、Western blot方法检测卵巢癌细胞和20对新鲜卵巢上皮癌组织及癌旁组织中CPT2 mRNA与蛋白的表达水平;另收集2010年1月至2014年8月就诊于西京医院的121例初治卵巢上皮癌患者的组织病理石蜡切片,运用免疫组织化学方法检测121例初治卵巢上皮癌组织石蜡病理切片中CPT2的表达,并分析CPT2与患者临床病理参数及生存预后的关系。结果与正常卵巢上皮细胞和癌旁组织相比,卵巢癌细胞和卵巢上皮癌组织中CPT2 mRNA和蛋白水平表达均显著下调,差异均有统计学意义(mRNA:t值分别为4.381、5.380,P<0.05;蛋白:t值分别为10.258、23.915,P<0.05)。免疫组化结果显示,卵巢上皮癌组织中CPT2的表达水平显著下调,并且CPT2低表达与患者Ⅱ型、Ⅲ~Ⅳ期、低分化、有网膜转移和淋巴结转移均有关联,差异均有统计学意义(χ^(2)值分别为10.140、18.812、20.221、4.211、17.405,P<0.05)。生存预后分析显示,CPT2的低表达与患者的不良预后显著关联(HR=0.790,95%CI:0.650~0.950,P<0.05)。结论卵巢癌细胞和卵巢上皮癌组织中CPT2表达显著下调,并与患者的不良预后有关,提示CPT2低表达在卵巢癌的发生发展中可能发挥促癌作用。
Objective To investigate the expression and clinical significance of carnitine palmitoyl transferase-2(CPT2)in ovarian epithelial cancer tissues,and to provide a new target for studying the treatment of ovarian cancer.Methods Fresh tissue specimens from 20 patients with epithelial ovarian cancer who were treated in Xijing Hospital from March 2015 to December 2016 were collected.The quantitative reverse transcription polymerase chain reaction(qRT-PCR)and Western blot were used to detect the expression levels of CPT2 mRNA and protein in ovarian cancer cells,20 pairs of ovarian epithelial cancer tissues and adjacent tissues.The histopathological paraffin sections of 121 patients with newly treated ovarian epithelial cancer who were treated in Xijing Hospital from January 2010 to August 2014 were collected.The expression of CPT2 in pathological sections of 121 newly diagnosed ovarian epithelial cancer tissues was detected by immunohistochemistry.The relationship between CPT2 expression and clinical significances and survival prognosis were analyzed.Results Compared with normal ovarian epithelial cells and adjacent tissues,the mRNA and protein expression levels of CPT2 in ovarian cancer cells and epithelial cancer tissues were significantly decreased,and the difference was statistically significant(mRNA:t=4.381 and 5.380,respectively,P<0.05;protein:t=10.258 and 23.915,respectively,P<0.05).The immunohistochemical results showed that the expression level of CPT2 in epithelial cancer tissues was significantly down-regulated,and low expression of CPT2 was correlated with typeⅡ,stageⅢ-Ⅳ,poorly differentiated,lymph node metastasis and momentum metastasis,and the difference was statistically significant(χ^(2)=10.140,18.812,20.221,4.211 and 17.405,respectively,P<0.05).Survival prognosis analysis also showed that the low expression of CPT2 was significantly correlated with the poor prognosis of patients,(HR=0.790,95%CI:0.650-0.950,P<0.05).Conclusion The expression of CPT2 in ovarian cancer cells and epithelial cancer tissues were significantly down-regulated and was related to the poor prognosis of patients,suggesting that the low expression of CPT2 may play a role in promoting the development of ovarian cancer.
