摘要
目的通过探讨大黄素对人精子中孕酮诱导的精子胞内钙离子浓度和精子特异性钙离子通道CatSper的影响,以期阐明大黄素抑制人精子功能的作用机制。方法选取2019年7月~2020年12月由江西省人类精子库提供的健康志愿者精液标本,通过钙信号测量技术和精子膜片钳技术分别检测人精子中孕酮诱导的精子胞内钙信号和CatSper电流变化。结果大黄素处理组与空白对照组相比,孕酮诱导的精子胞内钙信号增加幅度显著降低(P<0.05);大黄素处理组精子的CatSper电流以及孕酮激活的CatSper电流均显著低于对照组(P<0.05)。结论大黄素通过干扰人精子特异性钙离子通道CatSper抑制钙离子内流,进而抑制人精子功能。本研究对大黄素的临床应用具有一定的指导意义。
Objective To investigate the effect of emodin on progesterone-induced intracellular calcium ion concentration in human sperm and sperm-specific calcium channel CatSper,thus elucidating the mechanism through which emodin inhibits human sperm functions.Methods Semen specimens from healthy volunteers were collected from Jiangxi Human Sperm Bank from July 2019 to December 2020.The changes of intracellular calcium signal and CatSper current induced by progesterone in human sperm were detected through calcium signal measurement and sperm patch clamping,respectively.Results Compared with the control group,the increase magnitude of intracellular calcium signal in sperm induced by progesterone decrease significantly in emodin-treated group(P<0.05).The CatSper current and progesterone-activated CatSper current in human sperm are also significantly lower in the treatment group(P<0.05).Conclusion Emodin inhibits calcium influx by interfering with the specific calcium channel CatSper in human sperm,and thus restrains human sperm functions.This study provides certain guidance for the clinical application of emodin.
