外源药剂预处理对蒙古黄芪种子活力及活性氧、线粒体膜电位的影响 被引量:2

Effects of Exogenous Medicament Pretreatment on Seed Vigor,Active Oxygen and Mitochondrial Membrane Potential of Astragalus mongolica

摘要 为研究蒙古黄芪(Astragalus mongholicus Bunge)种子老化过程中活性氧和线粒体膜电位的变化与种子活力的关系,筛选提高种子抗老化能力外源药剂的适宜浓度,以不同浓度NAC和CsA预处理的种子为试材进行人工老化3 d处理后测定种子发芽率、发芽指数、活性氧含量和线粒体膜电位。实验结果表明,相比初始种子,蒙古黄芪种子经人工老化3d后,其胚轴活性氧含量升高、线粒体膜电位下降发芽率和发芽指数分别下降32.7%和40.6%;而经10 mmol/LNAC和90μmol/LCsA预处理的种子,相比于未进行外源药剂预处理(蒸馏水处理)的种子,在人工老化3d后,其发芽率分别提高31.2%和16.2%,发芽指数分别提高38.3%和21.1%,且胚轴活性氧含量有所下降,线粒体膜电位升高。表明通过适宜浓度的NAC和CsA预处理可以增强蒙古黄芪种子的抗老化能力,提高种子活力延长种子的寿命。 In order to study the relationship between the changes of active oxygen and mitochondrial membrane potential and seed vigor during the aging of Astragalus mongholicus Bunge seeds,the concentration of exogenous agents which was suitable for improving the anti-aging ability of seeds was screened.The seeds pretreated with different concentrations of NAC and CsA were used as the test materials,and after artificial aging for 3 days,the seed germination rate,germination index,active oxygen content and mitochondrial membrane potential were measured.The results showed that compared with the initial seeds,after artificial aging of A.mongholicus seeds for 3 days,the active oxygen content of hypocotyls increased,the mitochondrial membrane potential decreased,and the germination rate and germination index decreased by 32.7%and 40.6%,respectively.When the seeds were pretreated with 10 mmol/L NAC and 90μmol/L CsA,their germination rate increased by 31.2%and 16.2%,and germination index increased by 38.3%and 21.1%respectively compared with the seeds without exogenous agent pretreatment(distilled water treatment)after artificial aged for 3 days.Additionally,the hypocotyl active oxygen content decreased,and the mitochondrial membrane potential increased in exogenous agent treated seeds.It shows that pretreatment with appropriate concentrations of NAC and CsA can enhance the anti-aging ability of A.mongholicus seeds,increase seed vigor,and prolong seed life.
