摘要
本研究基于单因素法优选吸附率和解析率均较高的适于工业分离纯化苦参碱的大孔树脂及方法。利用高效液相色谱进行含量检测,通过静态和动态吸附与解析相结合的方法研究常用7种不同类型大孔吸附树脂对苦参提取液中苦参碱分离效果,优选最佳分离树脂及主要工艺参数。结果显示,HPD722分离效果最好动态吸附量为1.76 mg/mL树脂,最适分离条件为上样速度3 BV/h,上样溶液pH 9,70%乙醇洗脱流速3 BV/h、用量6 BV。从而得知HPD722可优选作为工业分离苦参碱的大孔吸附树脂,吸附量大,解析率高。
This study selected the suitable macroporous resins and methods to separate and purify matrine by high adsorption capacity and desorption rate.The content of samples was measured by the HPLC method.Combining static and dynamic adsorption and desorption experiments,the best resin was selected from 7 macroporous resins for matrine purification from Sophora flavescens.Results showed that HPD722 resin had the best performance with a adsorption capacity of 1.76 mg/mL resin.The optimal separation conditions were as follows sample loading speed was 3 BV/h,sample pH was 9,the flow rate of 70%ethanol elution was 3 BV/h,and the amount of 70%ethanol elution was 6 BV.As a result,HPD722 resin could be chosen to purify matrine for industrial production with the large adsorption capacity and high desorption rate.
