白腐真菌利用小麦秸秆产漆酶的筛选及产酶条件的优化 被引量:5

摘要 试验对9种白腐真菌进行筛选,获得利用小麦秸秆产漆酶水解圈较大的菌株,以小麦秸秆为碳源进行产漆酶液体发酵的单因素(培养时间、培养温度、初始pH值、碳源添加量、氮源种类、氮源浓度)试验和正交试验。研究表明,试验筛选出的高产漆酶的菌株为一色齿毛菌,对一色齿毛菌产漆酶进行单因素试验和正交试验,获得小麦秸秆的最佳添加量为25 g/L,最佳氮源为酵母浸粉,最佳氮源浓度为1.5 g/L,最佳培养基初始pH值为3.0,最佳培养温度为28℃。一色齿毛菌产漆酶的酶活由最初的1.09 U/mL提高至7.94 U/mL,较优化前提高7.28倍。 Nine kinds of white rot fungi were screened in the experiment,and the strains which produced laccase hydrolysis circle from wheat straw were selected.The single factor(culture time,culture temperature,initial pH value,carbon source concentration,nitrogen source type and nitrogen source concentration)and orthogonal experiments were carried out for liquid fermentation of laccase production with wheat straw as carbon source.The results showed that the high-yielding laccase strain screened by the experiment was Cerrena unicolor.The single factor test and orthogonal test were carried out on Cerrena unicolor.The optimal addition amount of wheat straw was 25 g/L,the best nitrogen source was yeast extract powder,the best nitrogen source concentration was 1.5 g/L,the optimal initial pH value of culture medium was 3.0,and the optimal culture temperature was 28℃.The laccase production activity of Cerrena unicolor increased from 1.09 to 7.94 U/mL,which was 7.28 times higher than that before optimization.
