钠钾ATP酶介导的腔上皮吸收与中医学收敛固涩作用的相关性研究

Study on the relationship between the absorption of luminal epithelium mediated by Na+,K+-ATPase and the astringency of traditional Chinese medicine

摘要 目的:探究钠钾ATP酶介导的腔上皮吸收与中医学收敛固涩作用的相关性。方法:通过功能类比研究钠钾ATP酶介导的腔上皮细胞吸收机制与中医学津液代谢的相关性,进一步通过实验研究收敛固涩类中药对正常与炎性状态下HCT-116细胞膜钠钾ATP酶活性的影响。结果:通过功能类比发现钠钾ATP酶介导的腔上皮细胞吸收机制与中医学的津液代谢具有很好的相关性,提出钠钾ATP酶很可能是收敛固涩类中药发挥固涩作用的重要靶蛋白的科学假说。进一步的实验研究发现收敛固涩类中药均能显著升高正常与炎性状态下HCT-116细胞膜钠钾ATP酶活性,而非收敛固涩类中药无该作用。结论:钠钾ATP酶是固涩类中药发挥收敛作用的关键蛋白。 Objective:To explore the relationship between Na+,K+-ATPase-mediated luminal epithelial absorption and the astringency of traditional Chinese medicine.Methods:Through functional analogy,the correlation between the absorption mechanism of Na+,K+-ATPase-mediated luminal epithelial cells and the theory of fluids was studied.Further experiments were carried out to study the effect of astringent Chinese medicines on HCT-116 cell membrane under normal and inflammatory conditions.Results:It was found that the absorption mechanism of luminal epithelial cells mediated by Na+,K+-ATPase had a good correlation with the body fluid metabolism of traditional medicine by functional analogy.It was proposed that Na+,K+-ATPase was likely to be an important target protein for astringent.Further experimental studies had found that astringent Chinese medicines could significantly increase the Na+,K+-ATPase activity in HCT-116 cell membrane under normal and inflammatory conditions,but non-astringent traditional Chinese medicines had no such effect.Conclusion:Na+,K+-ATPase is a key protein for astringent traditional Chinese medicines.
