摘要
目的:基于传统散剂和炒炙法探讨雷公藤的炮制减毒作用。方法:80只SPF级雄性小鼠随机分为正常组、生品组、清炒组、醋炙组、绿豆炙组、白芍炙组、甘草炙组、金钱草炙组,每组10只,各组连续28d灌胃给予雷公藤生品及各炮制品2g/kg,正常组给予等体积的溶媒,通过测定各组小鼠血清肝功指标丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST),肾功能指标肌酐(Cr)、尿素氮(BUN),评价其炮制减毒作用;通过对毒性靶器官肝和肾的丙二醛(MDA)及抗氧化指标超氧化物歧化酶(T-SOD)、微量还原型谷胱甘肽(GSH)、谷胱甘肽过氧化物酶(GPx)、谷胱甘肽-S转移酶(GST)水平的比较分析,初步探讨炮制减毒机制。结果:雷公藤引起了小鼠血清肝功和肾功指标的显著升高(P<0.01),肝和肾的脂质过氧化损伤(P<0.01)和抗氧化防御能力低下(P<0.01);而经醋炙和绿豆、白芍、甘草、金钱草分别煎汤炒炙后均可显著逆转血清肝功和肾功的异常(P<0.05,P<0.01),以及肝和肾的脂质过氧化损伤和抗氧化水平的低下(P<0.05,P<0.01)。结论:醋炙法和绿豆、白芍、甘草、金钱草煎汤炒炙法均对雷公藤的肝、肾毒性具有炮制减毒作用,其减毒机制可能涉及调控靶器官的氧化/抗氧化失衡。
Objective:To explore the detoxification effect of Tripterygium wilfordii Hook.f.based on traditional powder and stir-fried method.Methods:Eighty male SPF mice were randomly divided into normal group,raw product group,stirfried group,vinegar roasted group,10 in each group,Vigna radiata roasted group,Paeonia lactiflora roasted group,Glycyrrhiza uralensis roasted group,and Lysimachia christinae roasted group.Each group was given the Tripterygium wilfordii Hook.f.raw and processed products by gavage for 28 consecutive days at the dose of 2 g/kg,the normal group was given an equal volume of solvent.The serum liver function indexes alanine aminotransferase(ALT),aspartate aminotransferase(AST),renal function indexes creatinine(Cr),and urea nitrogen(BUN)of mice in each group were measured to evaluate its detoxification effect;and the mechanism was preliminarily discussed by the comparative analysis of the levels of lipid peroxidation(MDA)and antioxidant indexes superoxide dismutase(T-SOD),reduced glutathione(GSH),glutathione peroxidase(GPx)and glutathione-s transferase(GST)in liver and kidney of toxic target organs.Results:Tripterygium wilfordii Hook.f.caused a significant increase in serum liver and kidney function(P<0.01),lipid peroxidation damage of liver and kidney(P<0.01)and low antioxidant defense ability(P<0.01);while after being fried by stir-baking with vinegar,Vigna radiata,Paeonia lactiflora,Glycyrrhiza uralensis and Lysimachia christinae,the abnormalities of the above serum liver and kidney function were significantly reversed(P<0.05,P<0.01),as well as the lipid peroxidation damage and low antioxidant levels of liver and kidney(P<0.05,P<0.01).Conclusion:The vinegar broiling method and Vigna radiata,Paeonia lactiflora,Glycyrrhiza uralensis,and Lysimachia christinae decoction fried broiling method all could reduce the liver and kidney toxicity of Tripterygium wilfordii Hook.f.,and the detoxification mechanism may involve the regulation of the imbalance of oxidation and antioxidation in target organs.
