摘要
目的:建立全自动氨基酸分析仪测定复方α-酮酸片中5种游离氨基酸含量的方法,为其质量控制和质量标准的提高提供依据。方法:采用锂系统阳离子交换色谱柱和特定的柱后衍生化方法,以外标法同时测定复方α-酮酸片中5种氨基酸的含量。结果:复方α-酮酸片中的5种游离氨基酸分离度良好,空白无干扰。醋酸赖氨酸、苏氨酸、色氨酸、组氨酸、酪氨酸分别在5.31~53.26mg·L^(-1)(r=0.9998)、2.93~29.31mg·L^(-1)(r=0.9998)、1.22~12.20mg·L^(-1)(r=1.0000)、1.93~19.32mg·L^(-1)(r=0.9999)、1.50~15.04mg·L^(-1)(r=0.9998)范围内呈良好的线性关系。平均加样回收率(n=9)在98.2%~102.9%,RSD均不超过2.0%。结论:该方法简便、快速、准确,具有良好的重复性和稳定性,可用于复方α-酮酸片的质量控制。
Anion exchange chromatography column was used for amino acid analyzer(AAA)method with ninhydrin post column derivation.The results showed that good separation of free amino acids was achieved by this method with good linear relationship.The linear ranges were 5.31 mg·L^(-1)-53.26 mg·L^(-1)(r=0.9998),2.93mg·L^(-1)-29.31 mg·L^(-1)(r=0.9998),1.22mg·L^(-1)-12.20 mg·L^(-1)(r=1.0000),1.93mg·L^(-1)-19.32 mg·L^(-1)(r=0.9999),1.50mg·L^(-1)-15.04 mg·L^(-1)(r=0.9998)for LYS,THR,TRY,HIS,TYR,respectively.The average recoveries of five amino acids were within 98.2%-102.9%(n=9),the relative standard deviations were not more than 2.0%.The method is simple,rapid and accurate,which is suitable for quality control of compoundα-keto acid tablets.
