摘要
目的:为了解决离散细胞在透射电镜样品制备过程中细胞数量和超微结构保存困难的问题,探讨用蛋白(蛋清)对其进行预包埋的制备方法。方法:将离散的骨髓细胞和平滑肌细胞,分别予以蛋清预包埋和未预包埋方法进行处理,遂按透射电镜样品流程制备,而后镜下观察。结果:未预包埋组,两组细胞大多已流失,数量极少,无法包埋聚合。预包埋组,两组细胞呈现集中分布、数量可观,电镜观察均显示细胞超微结构清晰完整,细胞周围虽有微量的蛋白纤维,但对细胞结构观察无干扰。结论:在透射电镜样品制备过程中,对微量且离散的细胞样品使用蛋清进行预包埋处理是一种简便且有效的制备方法。
In order to solve the problem of difficult preservation of quantity and ultrastructure of easily dispersed cells in the process of preparation of transmission electron microscope samples,the method of pre-embedding them with albumen was discussed.Discrete bone marrow cells and smooth muscle cells were treated with albumen pre-embedding and non-embedding methods respectively,and prepared according to the sample flow of transmission electron microscope,then observed under the microscope.In the non-embedded group,most of the cells in the two groups were lost,and the number was very small,so they could not be embedded and polymerized.In the pre-embedding group,the cells in the two groups showed centralized distribution and considerable number.Electron microscopic observation showed that the cell ultrastructure was clear and complete.Although there were a small amount of protein fibers around the cells,there was no interference to the observation of cell structure.In the process of TEM sample preparation,it is a simple and effective method to embed micro and discrete cell samples with albumen.
