缺氧对肿瘤微环境的影响及其针对性治疗策略 被引量:2

摘要 肿瘤是对人类健康威胁最大的疾病之一。快速的细胞增殖和活跃的能量代谢是肿瘤的重要生物学特征。对于实体瘤,由于局部空间的限制和滞后的血管生成引起的血液灌流不足,极易导致肿瘤局部形成缺氧微环境。缺氧微环境在一定程度上可以通过抑制肿瘤的能量代谢而抑制其生长。然而对于部分恶性实体瘤,缺氧一方面可以刺激肿瘤细胞自身代谢及恶性生物行为学的改变,以适应乏氧微环境,另一方面将导致肿瘤细胞释放生物分子和生物信号对微环境进行改造,形成适瘤环境。已有研究表明,缺氧微环境是肿瘤发生、恶性进展、治疗耐受以及免疫逃逸的关键因素,因此,改善肿瘤微环境缺氧状态和阻断肿瘤细胞缺氧应答通路的治疗能提高肿瘤的治疗效果,开发相应的治疗药物和技术具有重要意义。
