运脾豁痰通脑方对脑缺血再灌注后胃肠损伤大鼠神经和胃肠功能、生长激素释放肽和小窝蛋白1表达的影响

摘要 目的分析运脾豁痰通脑方(自拟方)对脑缺血再灌注后胃肠损伤大鼠神经和胃肠功能、生长激素释放肽(Ghrelin)、小窝蛋白1表达的影响。方法将90只大鼠随机分为假手术组(30只)和缺血再灌注后胃肠损伤组(60只)。缺血再灌注后胃肠损伤组大鼠在大脑中动脉缺血再灌注24 h后均出现胃肠损伤表现为造模成功,将其再随机分为模型组和中药组各30只。造模成功3 d后,给予中药组灌胃运脾豁痰通脑方混悬液,其他两组灌胃蒸馏水。灌胃期间记录各组大鼠每日饮食量、腹泻的次数,并记录灌胃结束后的体重。灌胃14 d后,对各组大鼠进行神经功能缺损评分,处死大鼠后取其胃组织、脑组织分别用于检测Ghrelin、小窝蛋白1表达水平。结果(1)假手术组、中药组、模型组大鼠的体重依次降低、每日饮食量依次减少、每日腹泻次数依次增加(均P<0.05)。(2)假手术组大鼠无神经功能缺损,假手术组、中药组、模型组大鼠的神经功能缺损评分依次增加(均P<0.05)。(3)模型组和中药组的的小窝蛋白1和Ghrelin表达水平均高于假手术组(均P<0.05);中药组的小窝蛋白1表达水平低于模型组,而Ghrelin相对表达水平高于模型组(均P<0.05)。结论运脾豁痰通脑方不仅可以减轻脑缺血再灌注后胃肠损伤大鼠的神经功能损伤,还可改善胃肠道功能,其可能通过下调小窝蛋白1、上调Ghrelin的表达来发挥治疗作用。 Objective To analyze the effect of Yunpi Huotan Tongnao Fang(a self-designed formula)on neurological and gastrointestinal functions,growth hormone-releasing peptide(Ghrelin)and Caveolin 1 expressions in rats with gastrointestinal injury after cerebral ischemia-reperfusion.Methods Ninety rats were randomized to sham-operation group(n=30)or gastrointestinal injury after cerebral ischemia-reperfusion group(n=60).Rats in the gastrointestinal injury after cerebral ischemia-reperfusion group suffering from gastrointestinal injury after 24 hours of middle cerebral artery ischemia-reperfusion demonstrated successful modeling,then were randomly divided into model group(n=30)and Chinese medicine group(n=30).Three days after successful modeling,the Chinese medicine group was treated with suspension of Yunpi Huotan Tongnao Fang by intragastric gavage,and the remaining two groups with distilled water by intragastric gavage.For the rats of all groups,the daily food intake and frequency of diarrhea during intragastric gavage,and the weight at the end of intragastric gavage were recorded.After 14 days of intragastric gavage,the neurological function defect scoring was conducted on the rats in each group,then the rats were sacrificed to obtain their gastric tissues and brain tissues for the detection of Ghrelin and Caveolin 1 expression levels,respectively.Results(1)The weight and daily food intake decreased,and daily frequency of diarrhea increased in the order of the sham-operation group,the Chinese medicine group and the model group(all P<0.05).(2)No neurological function defect occurred in the rats of the sham-operation group,and the score of neurological function defect increased in the sham-operation group,Chinese medicine group,and model group successively(all P<0.05).(3)The expression levels of Caveolin 1 and Ghrelin in the model group and Chinese medicine group were higher than those in the sham-operation group(all P<0.05);compared with the model group,the Chinese medicine group obtained a lower Caveolin 1 expression level and a higher relative expression level of Ghrelin(all P<0.05).Conclusion Yunpi Huotan Tongnao Fang can not only alleviate the neurological function defect of rats with gastrointestinal injury after cerebral ischemia-reperfusion,but also improve the gastrointestinal functions.It exerts the therapeutic effect possibly by down-regulating Caveolin 1 expression and up-regulating Ghrelin expression.
