铁死亡相关基因对肝细胞癌患者诊断及预后预测价值的研究 被引量:1

摘要 目的探讨铁死亡相关基因对肝细胞癌(HCC)患者诊断及预后预测的价值。方法从TCGA数据库获取HCC转录组数据和临床资料,采用R软件“Limma”包筛选差异表达基因,并与FerrDb数据库的铁死亡相关基因取交集,得到铁死亡相关差异表达基因。进一步采用Kaplan-Meier生存分析筛选预后相关铁死亡相关差异表达基因,GO和KEGG进行功能富集分析。采用LASSO回归分析和Cox回归分析筛选关键预后相关的铁死亡相关差异表达基因。ROC曲线分析基因在HCC诊断及预后预测中的准确性。结果差异表达分析和生存分析筛选出ALB、CDKN2A、HRAS、JUN、MAPK3、NOX4和SRC这7个预后相关的铁死亡相关差异表达基因。功能富集分析结果显示,这些基因主要为结合蛋白,富集在ErbB信号通路、GnRH信号通路、VEGF信号通路、AGE-RAGE信号通路以及线粒体自噬等通路中。LASSO回归分析和Cox回归分析结果显示,SRC可作为HCC的独立预后因子(P<0.05)。ROC曲线显示,SRC在HCC诊断和预后预测上具有较高的准确性。结论铁死亡抑制因子SRC在HCC患者的诊断及预后预测中具有较好的价值,是一个潜在的癌症治疗靶点。 Objective To explore the diagnostic and prognostic value of ferroptosis-related genes in patients with hepatocellular carcinoma(HCC).Methods The transcriptome data and clinical data of HCC were obtained from the TCGA database,R software"Limma"package was used to screen differentially expressed genes.The genes were taken the intersection with the ferroptosis-related genes from the FerrDb database,which obtained ferroptosis-related differentially expressed genes.Kaplan-Meier survival analysis was further used to screen the prognostic-related ferroptosis-related differentially expressed genes,GO and KEGG were employed to conduct functional enrichment analysis.LASSO regression analysis and Cox regression analysis were used to screen the key prognostic related ferroptosis-related differentially expressed genes.The accuracy of genes in the diagnosis and prognosis of prediction HCC were analyzed by ROC curve.Results Differential expression analysis and survival analysis screened out seven prognostic-related ferroptosis-related differentially expressed genes,including ALB,CDKN2A,HRAS,JUN,MAPK3,NOX4 and SRC.Functional enrichment analysis result showed that these genes were mainly binding proteins,which were enriched in ErbB signaling pathway,GnRH signaling pathway,VEGF signaling pathway,AGE-RAGE signaling pathway and mitochondrial autophagy.LASSO regression analysis and Cox regression analysis results showed that SRC could be used as an independent prognostic factor for HCC(P<0.05).ROC curve indicated that SRC had a higher accuracy in HCC diagnosis and prognosis prediction.Conclusion The ferroptosis suppressor SRC has a preferable value in the diagnosis and prognosis prediction of HCC patients,which is a promising therapeutic target for cancer.
