摘要
目的探讨长链非编码RNA(lncRNA)同源盒基因A11反义RNA(HOXA11-AS)在急性淋巴细胞白血病(ALL)患儿骨髓中的表达及意义。方法选取108例初治ALL患儿及57例非肿瘤性疾病患儿,比较ALL患儿与非肿瘤性疾病患儿、不同ALL病理类型、治疗后不同疗效ALL患儿HOXA11-AS的相对表达量。实时逆转录聚合酶链反应(RT-PCR)检测108例不同临床特征ALL患儿骨髓中HOXA11-AS的相对表达量。结果108例初治ALL患儿骨髓HOXA11-AS的相对表达量明显高于非肿瘤性疾病患儿(P﹤0.01),其中,B-ALL型初治ALL患儿骨髓HOXA11-AS的相对表达量明显低于T-ALL型患儿(P﹤0.01)。108例初治ALL患儿经2个周期的化疗后完全缓解46例,未完全缓解62例,完全缓解患儿HOXA11-AS的相对表达量明显低于未完全缓解和初治ALL患儿(P﹤0.01)。BCR/ABL融合基因阳性表达初治ALL患儿骨髓HOXA11-AS的相对表达量,明显高于BCR/ABL融合基因阴性表达患儿(P﹤0.05)。结论HOXA11-AS在ALL患儿中表达上调,并与ALL分型有关,其可能参与ALL的发生发展过程。
Objective To explore the expression and significance of long noncoding RNA(lncRNA)homeobox A11 antisense RNA(HOXA11-AS)in the bone marrow of children with acute lymphoblastic leukemia(ALL).Method A total of 108 cases of newly-diagnosed ALL children and 57 children with non-neoplastic diseases were selected.Compare the relative expression of HOXA11-AS in children with ALL and children with non-neoplastic diseases,different pathological types of ALL,and different curative effects after treatment.Real-time reverse transcription polymerase chain reaction(RT-PCR)was used to detect the relative expression of HOXA11-AS in the bone marrow of 108 children with ALL with different clinical characteristics.Result The relative expression of HOXA11-AS in the bone marrow of 108 newlydiagnosed ALL children was significantly higher than that in children with non-neoplastic diseases(P<0.01).Among them,the relative expression of HOXA11-AS in the bone marrow of children with B-ALL newly-diagnosed ALL was significantly lower than that of children with T-ALL(P<0.01).After 2 cycles of chemotherapy,108 cases of newly newly-diagnosed ALL children were a complete response to the therapy,and 46 cases were partial response,and The relative expression of HOXA11-AS in children with complete remission was significantly lower than that in children with incomplete remission and newly-treated ALL(P<0.01).The relative expression of HOXA11-AS in the bone marrow of newlytreated ALL children with BCR/ABL fusion gene-positive was significantly higher than that of children with fusion gene negative,the difference was statistically significant(P<0.05).Conclusion HOXA11-AS is up-regulation in children with ALL and is related to ALL classification,which may be involved in the initial and development of ALL.
