青海高山辣根菜中水溶性总多糖的提取工艺研究 被引量:4

摘要 探讨青海高山辣根菜中水溶性总多糖的最佳提取工艺,优化定量方法。以高山辣根菜中水溶性总多糖的提取率为指标,以提取时间、提取次数、提取温度、固液比为考察因素,采用苯酚—硫酸法测定多糖含量。利用单因素实验确定提取因素水平,通过正交设计方法,优选高山辣根菜中多糖的最佳提取工艺。结果表明,最佳提取工艺为:固液比1∶20,提取1次,提取时间2.5h,提取温度85℃。通过优选出的最佳提取工艺条件所得总多糖含量高,确定高山辣根菜中总多糖的提取工艺简单有效可行。 To study the optimal extraction technology and optimized quantitative method of water soluble total polysaccharide from Pegaeophyton scapiflorum in Qinghai.Taking the extraction rate of water soluble total polysaccharide in Pegaeophyton scapiflorum as index,and using the extraction time,extraction times,extraction temperature and solid-liquid ratio as investigate factors,the content of total polysaccharide was determined by phenol-sulfuric acid method.Single factor experiment was used to determine the level of extraction factors,and orthogonal design method was used to optimize the extraction process of water soluble total polysaccharide from Pegaeophyton scapiflorum.The results showed that the optimal extraction conditions were as follows:solid-liquid ratio of 1∶20,extract time of 1,extraction time of 2.5 h,extraction temperature of 85℃.Though the optimal extraction conditions,the content of total polysaccharide was high,and the extraction process of total polysaccharide from Pegaeophyton scapiflorum was simple,effective and feasible.
