血红素基因hemA和hemC对链霉菌HCCB10043产A21978C的影响

摘要 目的考察血红素合成基因的异源表达对链霉菌HCCB10043产环脂肽类抗生素A21978C的影响。方法应用PCR方法扩增血红素合成基因hemA、hemC,使用属间接合转移的方法转化入链霉菌HCCB10043中,获得异源表达菌株。通过HPLC检测突变株A21978C产量的变化。结果通过筛选获得血红素合成基因hemA、hemC异源表达菌株,过表达后突变株A21978C产量相比亲株增加了20%。结论血红素合成基因的异源表达可提高链霉菌代谢产物A21978C的产量。 Objective To investigate the effects of heterologous expression of heme biosynthetic genes on the production of A21978C by Streptomyces HCCB 10043.Methods The genes hemA and hemC were amplified by PCR and transformed into the strain of Streptomyces HCCB 10043 by intergeneric conjugation.The contents of A21978C produced by the mutant strains were detected by HPLC.Result The strains for the heterologous expression of hemA and hemC were screened and the production of A21978C was increased by 20%compared with the original strain.Conclusion Heterologous expression of heme biosynthetic genes can increase the A21978C production of Streptomyces.
