苹果半矮化砧木G.935的离体快繁体系建立 被引量:3

摘要 以苹果优良半矮化砧木G.935为试材,以当年生半木质化新梢芽段为外植体,用次氯酸钠表面杀菌后置腋芽启动培养基上诱导腋芽萌发生长形成无菌绿苗;以健壮无菌绿苗为试材,设置不同的基本培养基和外源激素种类及浓度,以筛选适宜无菌苗增殖培养和不定根诱导的培养基。结果表明,MS+0.5 mg·L^(-1)BA+0.1 mg·L^(-1)IBA是G.935无菌苗增殖的最佳培养基。以基本培养基1/2MS+IBA进行生根诱导的生根率和平均单株生根数较高,尤其IBA浓度为0.2 mg·L^(-1)时效果最好,生根率可达98.3%,平均单株生根数达7条以上。本研究建立了砧木G.935从试管苗启动培养、增殖到生根的高效离体快繁体系。 The semi-lignified new growing shoots of the good apple semi-dwarfing rootstock G.935 were used as explants, and the stem section with one axillary bud after surface sterilized with sodium hypochlorite was explanted on medium to induce axillary bud break and growth. With the obtained in vitro shoots as materials, different basal media, kinds and concentrations of exogenous hormones were set to screen out the suitable media for mulitiplication culture and adventitious root induction. The results showed that the optimal mulitiplication medium was MS+0.5 mg·L^(-1)BA+0.1 mg·L^(-1)IBA. For root induction, higher rooting rate and average root number per plantlet were obtained on the medium 1/2 MS+IBA, especially when the IBA concentration was 0.2 mg·L^(-1), the rooting rate could reach 98.3%, and average root number per plantlet was more than 7. The high-efficiency in vitro rapid propagation system of G.935 was established including in vitro culture of tuber plantlets, mulitiplication culture and rooting culture.
