金属有机骨架-碳点纳米电化学发光体用于尿酸的灵敏检测 被引量:2

摘要 尿酸(Uric Acid,UA)含量是人体健康的一个重要指标,UA含量异常会导致肾结石、关节炎、心血管疾病,因此,人体血液中UA含量的检测具有重要意义。该工作制备了金属有机骨架固载的碳点复合纳米材料,并以此作为发光体构建了电化学发光生物传感器,用于UA的高灵敏检测。首先,以鲁米诺(Luminol)和二硫代二苯甲酸(DTSA)为原料,通过溶剂热法合成了硫原子掺杂的碳点(CDs)。然后在含有CDs的溶液中合成了金属有机骨架ZIF-8,通过原位生长包埋法使得大量CDs在ZIF-8生长成型的过程中被负载于ZIF-8框架上,合成了具有强ECL信号的CDs@ZIF-8复合材料。实验结果表明,UA能够有效猝灭CDs@ZIF-8/溶解氧体系的ECL信号,该传感器对UA响应的线性范围为5 nmol/L~100μmol/L,检测限达5.4 nmol/L。 The content of Uric Acid(UA)is an important indicator of human health.And the abnormal content of UA will lead to kidney stones,arthritis,cardiovascular diseases,etc.Therefore,the detection of UA content in human blood is of great significance.In this work,carbon dots embedded metal-organic framework nanocomposite were prepared and used as emitter to construct an electrochemiluminescence biosensor,which could be used for highly sensitive detection of UA.First,sulfur doped carbon dots(CDs)were synthesized by solvothermal method from luminol and dithiobenzoic acid(DTSA).Then,the metal organic framework ZIF-8 was synthesized in the solution containing CDs.By in-situ embedding,a large number of CDs were loading on ZIF-8 during the growth and molding process of ZIF-8,and the CDs@ZIF-8 nanocomposite with strong ECL signal was acheived.The results show that UA can effectively quench the ECL signal of CDs@ZIF-8/dissolved oxygen system.The linear range of the biosensor's response to UA is from 5 nmol/L to 100μmol/L,and the detection limit is up to 5.4 nmol/L.
