摘要
目的分析不同检测方法检测宫颈癌组织程序性死亡配体-1(programmed death ligand 1,PD-L1)和T细胞免疫球蛋白黏蛋白分子-3(T-cell immunoglobulin mucin 3,TIM-3)配体半乳糖凝集素-9(galectin-9)的一致性。方法选择宫颈癌患者100例,分别采用RNA原位杂交(RNA in situ-hybridization,RNAscope)、荧光原位杂交(fluorescence in situ hybridization,FISH)和免疫组织化学(immunohistochemistry,IHC)检测宫颈癌组织标本PD-L1和galectin-9的表达水平及一致性。结果采用RNAscope、FISH和IHC检测宫颈癌患者PD-L1表达水平的阳性率分别为71.0%、20.0%、77.0%,与FISH比较,RNAscope和IHC检测宫颈癌患者PD-L1表达水平较高(P<0.001),其中RNAscope和IHC检测结果Kappa=0.940,一致性好。采用RNAscope、FISH和IHC检测宫颈癌患者galectin-9表达水平的阳性率分别为72.0%、22.0%、75.0%,与FISH比较,RNAscope和IHC检测宫颈癌患者galectin-9表达水平较高(P<0.001),其中RNAscope和IHC检测结果Kappa=0.970,一致性好。结论采用RNAscope和IHC等病理技术检测宫颈癌特异性PD-L1和galectin-9的一致性高,为今后开展特异性免疫治疗宫颈癌患者提供了重要的依据。
Objective To analyze the consistency of different detection methods for detection of programmed death ligand 1(Programmed death ligand 1,PD-L1)and T-cell immunoglobulin and mucin domain-3(T-cell immunoglobulin mucin 3,TIM-3)ligand galectin-9(galectin-9)in cervical cancer tissue.Methods A total of 100 patients diagnosed with cervical cancer from January 2018 to January 2020 in the department of pathology of our hospital were selected.RNA in situ hybridization(RNAscope),fluorescence in situ hybridization(FISH)and immunohistochemistry(IHC)were used to detect the expression levels and consistency of PD-L1 and galectin-9 in cervical cancer tissue samples.Results The positive rates of PD-L1 expression in patients with cervical cancer detected by RNAscope,FISH and IHC were 71.0%,20.0%and 77.0%,respectively.Compared with FISH,the expression levels of PD-L1 in patients with cervical cancer detected by RNAscope and IHC were higher(P<0.001),and the results of RNAscope and IHC Kappa=0.940,showing good consistency.The positive rates of galectin-9 expression in patients with cervical cancer detected by RNAscope,FISH and IHC were 72.0%,22.0%and 75.0%,respectively.Compared with FISH,galectin-9 expression in patients detected by RNAscope and IHC was higher(P<0.001),and Kappa=0.970 of the results of RNAscope and IHC were consistent.Conclusion The detection of PD-L1 and galectin-9 by RNAscope and IHC was highly consistent,which provided an important basis for the specific immunotherapy of cervical cancer patients in the future.
