摘要
目的研究改良后制作方法建立大鼠大脑中动脉栓塞(MCAO)模型的成功率和存活率。方法选取健康清洁级雄性SD大鼠30只,使用自制石蜡线栓,采用异氟烷建立大鼠MCAO模型。通过形态学检测(TTC染色)、神经功能5分制评分法观察模型成功率,以及大鼠2、7 d内的存活率。结果符合神经功能5分制评分法标准即建模成功的大鼠28只,成功率为93.33%。模型制备后2、7 d内,存活率分别为100.00%(28/28)、82.14%(23/28)。结论采用自制石蜡线栓和异氟烷麻醉适合MCAO模型制备,存活率和成功率均较高,但在具体实践过程中仍需注意方法和技巧。
Objective To study success rate and survival rate of the improved manufacturing method to establish the middle cerebral artery occlusion(MCAO)model in rats.Methods Thirty healthy and clean-grade male SD rats were selected.The rat models of MCAO were established with isoflurane and self-made paraffin thread suppository.The success rate of model and the survival rate in 2,7 d were observed by morphological examination(TTC staining)and neurological function 5 points scoring system.Results Twenty-eight rats met the criteria of the 5-point neural function scoring method,namely,the modeling was successful,and the success rate was 93.33%.Within 2 and 7 days after model preparation,the survival rate was 100.00%(28/28),82.14%(23/28)respectively.Conclusion Using self-made paraffin suture and isoflurane anesthesia is suitable for MCAO model preparation,and the survival rate and success rate are both high,but methods and skills still need to be paid attention to in the specific practice process.
