HPLC法测定蒙成药萨仁-嘎日迪中羟基红花黄色素A的含量

Determination of Hydroxysafflor Yellow A in Mongolian Saren-Gridi by HPLC

目的:建立HPLC法测定蒙成药萨仁-嘎日迪中羟基红花黄色素A的含量。方法:采用InertSustain AQ-C_(18)(250 mm×4.6 mm,5μm)色谱柱,流动相为用三乙胺调至pH(6.0±0.1)的甲醇-乙腈-0.7%磷酸(19.5∶1.5∶79),流速为1.0 mL/min,检测波长403 nm,柱温40℃,进样量为10μL。结果:羟基红花黄色素A在0.03006~1.2024μg范围内呈良好的线性关系,相关系数为r=0.9999;平均回收率97.38%(RSD=0.88%)。结论:所建立的HPLC法稳定可靠,可作为萨仁-嘎日迪中羟基红花黄色素A含量测定的方法。

Objective To establish the high performance liquid chromatography(HPLC)for determination of Hydroxysafflor yellow A in Mongolian Saren-Gridi.Methods The HPLC was carried out with InertSustain AQ-C_(18)(250 mm×4.6 mm,5μm),Using methanol-acetonitrile-0.7%phosphoric acid solution(19.5∶1.5∶79)as mobile phase(PH(6.0±0.1)adjusted by triethylamine)at a flow rate of 1.0 mL·min^(-1).The wavelength was set at 403 nm,the column temperature was 40℃,and the injection volume was 10μL.Results The linear of hydroxysafflor yellow A was in the range of 0.03006μg~1.2024μg(r=0.9999).The average recovery was 97.38%(RSD=0.88%).Conclusion This method is simple,high accuracy and specificity.It can be used for the determination of hydroxysafflor yellow A.
