摘要
目的探究复方桑叶消痤凝胶对ICR小鼠痤疮模型的相关抗炎影响。方法将6~8周龄雌性ICR小鼠随机分为:生理盐水对照组、模型组、七味姜黄搽剂组、复方桑叶消痤凝胶高、中、低剂量治疗组,n=6。小鼠耳廓内注射P.acnes致炎24h,各组行生理盐水、七味姜黄搽剂、复方桑叶消痤凝胶高、中、低剂量涂抹治疗,持续治疗7 d。记录耳廓厚度及形态学变化,行常规HE染色,实时荧光定量PCR法检测组织NLRP3、caspase-1、IL-1β的mRNA表达水平,蛋白免疫印迹法检测TLR2等相关蛋白表达水平。结果复方桑叶消痤凝胶能对痤疮炎症起一定改善作用,减轻表皮脊层增生,皮损中炎症小体相关因子NLRP3、caspase-1、IL-1βmRNA表达降低,皮损中TLR2、NLRP3、caspase-1蛋白表达降低。结论复方桑叶消痤凝胶减轻了痤疮模型小鼠的炎症,这种炎症改善作用可能受NLRP3炎性小体信号通路调控。
Objective:To explore the anti-inflammatory effect of Compound Sangye Xiaocuo Gel on ICR mouse acne model.Methods:Female ICR mice aged 6-8 weeks were randomly divided into normal saline control group,model group,seven-flavored turmeric liniment group,Compound Sangye Xiaocuo Gel groups of high,medium and low doses,n=6.The mice were injected with P.acnes into the auricle to cause inflammation for 24 hours,and each group was treated with normal saline,seven-flavored turmeric liniment,Compound Sangye Xiaocuo Gel of high,medium and low doses,and the treatment lasted for 7 days.The thickness and morphological changes of the auricle were recorded,and conventional HE staining was performed.The mRNA expression levels of NLRP3,caspase-1 and IL-1βin tissues were detected by real-time fluorescent quantitative PCR,and the expression levels of TLR2 and other related proteins were detected by western blotting.Results:Compound Sangye Xiaocuo Gel can improve acne inflammation,reduce epidermal spine hyperplasia,and lower inflammasome-related factors of NLRP3,caspase-1,IL-1βmRNA expression in skin lesions and the protein expression of TLR2,NLRP3 and Caspase-1 in skin lesions.Conclusion:Compound Sangye Xiaocuo Gel has reduced the inflammation in acne model mice,and this inflammation improvement effect may be regulated by the NLRP3 inflammasome signaling pathway.
