摘要
目的初步探讨滇三七联合氟康唑抗耐药白念珠菌的体内外作用效果。方法用微量稀释法测定滇三七联合氟康唑对白念珠菌氟康唑耐药株的体外抑菌活性。建立耐药白念珠菌感染的外阴阴道念珠菌病(VVC)小鼠模型,通过阴道灌洗给药进行治疗,观察小鼠的全身状况、体重变化、外阴炎症评分及阴道灌洗液菌落数统计,PASM染色观察阴道组织病理学变化,从而评价滇三七联合氟康唑的体内抗菌作用。结果两药联合在体外对5株白念珠菌氟康唑耐药株均具有良好的协同抗菌作用。体内结果显示,联合组可改善VVC小鼠的一般状况,阴道局部炎症评分显著降低(P<0.05),阴道灌洗液菌落数显著减少(P<0.01),阴道组织病理切片中的白念珠菌明显减少。结论滇三七联合氟康唑在体内外均有良好的协同抗耐药白念珠菌的效果,能明显改善小鼠外阴阴道念珠菌病的症状。
Objective To investigate the antifungal activity of panax notoginseng combined with fluconazole on drug-resistant Candida albicans in vitro and in vivo.Methods The effect of panax notoginseng combined with fluconazole on Candida albicans resistant strains was determined by the microdilution method.The antifungal activity of panax notoginseng combined with fluconazole was also assessed in the murine vulvovaginal candidiasis(VVC) model.The mice were infected with drug-resistant Candida albicans strains.The general condition and weight of the mice were observed, the vaginal inflammation was scored, the number of vaginal lavage colonies was determined, and the pathological changes of vaginal tissue was observed by PASM staining.Results Panax notoginseng combined with fluconazole had a good synergistic effect on 5 fluconazole-resistant Candida albicans strains in vitro.The treatment also showed good effects in vivo.The general condition of VVC mice was improved, the vaginal local inflammation score was reduced(P<0.05), the number of colonies was decreased markedly in vaginal lavage(P<0.01), and the number of Candida albicans in vaginal tissue pathological sections was significantly reduced.Conclusion Panax notoginseng combined with fluconazole has synergistic therapeutic effect on resistant Candida albicans in vitro and in vivo, and can significantly improve the symptoms of vulvovaginal candidiasis in mice.
