水牛PADI6重组蛋白多克隆抗体的制备及表达模式分析

摘要 为了解PADI6母源基因在水牛上的表达模式,初步阐明母源基因对卵母细胞的成熟和胚胎早期发育的作用,首先分析了PADI6基因在水牛上的表达情况,通过基因克隆,制备多克隆抗体,发现PADI6基因在不同物种中进化具有保守性。将所得PADI6多克隆抗体进行特异性表达分析,发现PADI6仅在卵母细胞中表达。为进一步了解PADI6母源基因在早期胚胎的表达情况。对水牛PADI6母源基因转录水平进行了分析,发现转录产物在GV期和MⅡ期卵母细胞中大量积累,在桑椹胚和囊胚阶段显著降低。上述研究结果表明,母源基因PADI6对卵母细胞的成熟和胚胎早期发育至关重要。 The purpose of this study was to investigate the expression pattern of PADI6 maternal genes on buffalo,and to preliminarily clarify the effect of maternal gene on oocyte maturation and early embryonic development.Firstly,the expression pattern of PADI6 maternal genes was analyzed on buffalo,Through gene cloning,polyclonal antibodies were prepared,and it was found that PADI6 gene evolution was conserved in different species.The polyclonal antibody PADI6 was analyzed for specific expression,and it was found that PADI6 was only expressed in oocytes.In order to further understand the expression of PADI6 maternal gene in early embryos,the transcript levels of the buffalo PADI6 maternal gene were analyzed,and the transcript was found to be heavily accumulated at the GV and MⅡstage oocytes until it decreased in the 16-cell stage,and significantly decreased in the morula and blastocyst stages.These results suggest that the maternal gene PADI6 is important for oocyte maturation and early embryonic development.
