摘要
目的建立同时测定喉康散中靛蓝和靛玉红含量的高效液相色谱法。方法色谱柱为Phenomenex Gemini C_(18)柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.05 mol/L醋酸铵溶液(75∶25,V/V),流速为1.0 m L/min,检测波长为289 nm,柱温为30℃,进样量为10μL。结果靛蓝和靛玉红质量浓度分别在5.664~56.640μg/m L(r=0.9999)和0.4288~4.2880μg/m L(r=0.9992)范围内与峰面积线性关系良好;靛蓝、靛玉红平均回收率分别为96.28%和91.55%,RSD分别为1.06%和1.43%(n=6);精密度、重复性、稳定性试验结果的RSD均低于2.00%。结论该方法操作简便,结果准确可靠,专属性强,重复性和稳定性均较好,可用于同时测定喉康散中靛蓝和靛玉红的含量。
Objective To establish a high-performance liquid chromatography(HPLC)method for the simultaneous determination of indigotin and indirubin in Houkang Powder.Methods The chromatographic column was Phenomenex Gemini C_(18) column(250 mm×4.6 mm,5μm),the mobile phase was methanol-0.05 mol/L ammonium acetate solution(75∶25,V/V),the flow rate was 1.0 m L/min,the detection wavelength was 289 nm,the column temperature was 30℃,and the injection volume was 10μL.Results The linear ranges of indigotin and indirubin were 5.664-56.640μg/m L(r=0.9999)and 0.4288-4.2880μg/m L(r=0.9992),respectively.The average recoveries of indigotin and indirubin were 96.28%and 91.55%with RSDs of 1.06%and 1.43%(n=6),respectively.The RSDs of precision,repeatability and stability tests were lower than 2.00%.Conclusion The method is simple,accurate,reliable,specific,reproducible and stable,which can be used for the content determination of indigotin and indirubin in Houkang Powder.
