摘要 为优化颠茄草中硫酸阿托品的提取工艺,采用超声波提取硫酸阿托品,用乙酸乙酯萃取纯化,通过HPLC法测定其含量并探讨乙醇浓度、超声时间、料液比、粒度4个因素对提取率的影响,通过单因素和正交实验确定最优的提取工艺.研究结果表明,过50目筛的颠茄粉,以料液比1∶50的比例加入85%的乙醇溶液,超声提取30 min,提取率最高,其为0.61 mg/g. In order to optimize the extraction process of atropine sulfate from belladonna,the atropine sulfate was extracted by ultrasonic method,purified by ethyl acetate,and its content was determined by HPLC method.The influence of ethanol concentration,ultrasonic time,material-to-liquid ratio and particle size on the extraction rate were discussed.The optimal extraction process was determined by the single factor and orthogonal experiments.The results showed that the belladonna powder passed through a 50-mesh sieve was added with 85%ethanol solution at a ratio of 1∶50 to the material-to-liquid ratio,followed by ultrasonic extraction for 30 min,and the extraction rate was the highest with a content of 0.61 mg/g.
