臭氧水对小鼠急性毒性和蓄积毒性试验 被引量:2

Acute Toxicity and Accumulative Toxicity Tests of Ozone Water in Mice

摘要 用昆明种小白鼠作为实验动物分别进行急性毒性试验和蓄积毒性试验,来评价臭氧水的毒性。急性毒性试验采用最大给药法,试验组取浓度≥20 mg/kg的臭氧水按20 g/kg体质量限量,一次灌胃,观察14 d内小鼠的死亡情况,计算半数致死量(Median Lethal Dose,LD_(50))。蓄积毒性试验采用剂量递增法,按体质量计算染毒剂量。处理后1~4 d小鼠染毒剂量为0.1倍的LD_(50),按20 g/kg体质量,一次灌胃给药,以后每4 d递增1.5倍剂量,直至20 d。每4 d对所有小鼠进行称重,统计增重和采食量,并测定小鼠器官指数,观察胃组织切片。结果表明:试验期间无小鼠死亡,臭氧水对小鼠的经口LD_(50)为20 g/kg体质量,根据急性毒性分级,属实际无毒物质。臭氧水的蓄积系数K>5。与灌注纯净水的对照组小鼠相比,灌注臭氧水对小鼠无任何不良影响。 Kunming mice were used as experimental animals to conduct acute toxicity and accumulative toxicity tests to assess the potential toxicity of ozone water.The acute toxicity test of ozone water was carried out by using maximal dosage administration.The mice in test group were administrated ozone water with a concentration≥20 mg/kg according to 20 g/kg body mass limit.With one stomach gavage,the death of mice within 14 days was observed,and the median lethal dose(Median Lethal Dose,LD_(50))was calculated.Accumulative toxicity test of ozone water was carried out by dose escalation,and the exposure dose was calculated according to body mass of the mice.The mice were administrated 0.1 LD_(50) dosages during the first 4 days,and then dosages with 1.5 fold increase every 4 days until 20 days.All mice were weighted every 4 days,and the weight gain and feed intake were counted.Meanwhile,the organ indexes of the mice were measured and the stomach tissue sections were observed.The result showed that no mice died during the test and the oral LD_(50) of ozone water to mice was 20 g/kg body mass.According to the acute toxicity classification,it is a non-toxic substance.Accumulative test showed that accumulation coefficient was no less than 5.Compared with purified water,ozone water did not have any adverse effects on the mice.
