豌豆黄叶突变体转录组测序和表达模式分析 被引量:1

摘要 为了探究豌豆黄化突变体形成机制,以豌豆黄化幼苗突变体(yl)和野生型植株(WT)为材料,对2份材料的叶片进行比较转录组分析。结果显示,利用高通量转录组测序技术分别从WT和yl中分别获得25599903、24806761条片段;通过对2份材料差异基因的筛选,共得到4423个差异表达基因(DEGs),其中,2449个上调表达,1974个下调表达;对其中9个相关基因进行了RT-qPCR验证,表达趋势与RNA-Seq结果一致。GO功能富集分析发现,差异基因广泛涉及生物合成与代谢等途径。对参与叶绿素合成和光合作用途径的DEGs进行表达模式分析,发现叶绿素降解与合成途径的关键基因可能与yl的形成有关。 Understanding the molecular mechanisms of chloroplast development and chlorophyll biosynthesis in pea(Pisum sativum L.)is extremely important for pea breeding.This study used high-throughput transcriptome sequencing and bioinformatics analyses to examine changes in gene expression in leaves of yellow-leaf mutant(yl)and wild type(WT),and compared the gene expression profiles of these two pea types.A total of 25599903 and 24806761 high quality reads were obtained for the WT and the mutant,respectively.In total,4423 differentially expressed genes(DEGs)were identified in yellow-leaf mutant compared to WT,of which 2449 were up-regulated and 1974 were down-regulated.Gene ontology and pathway enrichment analyses indicated that the DEGs were involved in various biosynthetic and metabolic pathways.Expression profiles of genes involving chlorophyll synthesis and photosynthetic pathways were illustrated.Quantitative RT-PCR analysis of the relevant DEGs confirmed the RNA-Seq results.Expression patterns of DEGs related with Chl metabolism showed that the key genes of chlorophyll synthesis and photosynthetic pathways may be related to the formation of yl.
