摘要 目的不同人参的品质差异体现在细微之处,稀有人参皂苷Rg3正是一种重要的、微量的活性成分;本研究旨在建立人参Rg3的测定方法,为人参质量评价和鉴别提供依据。方法利用标准差分析,方法学考察(线性、精密度及准确度)建立了Rg3的液液萃取制样及其高效液相色谱含量测定的方法,并对野山参、红参及园参中的Rg3进行含量测定。结果高效液相色谱法测定S型Rg3的线性范围为0.04~0.20 mg.mL^(-1),相关系数r为0.9975,最低检测限为0.04 mg.mL^(-1),精密度(n=6)为1.21%;测定R型Rg3的线性范围为0.04~021mg.mL^(-1),相关系数r为0.9989,最低检测限为0.04 mg.mL^(-1),精密度(n=6)为1.12%;以野山参为样本,S型Rg3回收率为99.06%。在所检测的样本中,野山参只含有S型Rg3,而红参则同时检测出S型和R型两种构型的Rg3,园参未检出Rg3。结论高效液相色谱法测定人参皂苷Rg3灵敏准确,重现性好,适用于人参及其制品中Rg3的含量测定。 Objective The quality difference of various ginseng is reflected with a subtle material,but Ginsenoside Rg3 had been concerned as an important trace active ingredient in Ginseng,which was called a rare ginsenosides,thus the research had been made to establish a method for the determination of ginsenoside Rg3,and provide evidence for quality evaluation and identification of ginseng.Methods By means of standard deviation analysis and methodological investigation(linearity,precision and accuracy),the method of liquid-liquid extraction of Rg3 and its content determination by high performance liquid chromatography were established.Results the linear range of S-type Rg3 was 0.04~0.20mg.ml^(-1),the correlation coefficient r was 0.9975,the minimum detection limit was 0.04mg.ml^(-1),and the precision(n=6)was 1.21%.The linear range of the determination of R-type Rg3 was 0.04-0.21 mg.ml^(-1),the correlation coefficient r was 0.9989,the minimum detection limit was 0.04mg.ml^(-1),and the precision(n=6)was 1.12%;the recovery rate of s-type Rg3 in wild ginseng was 99.06%.In the tested samples,wild ginseng only contained s-type Rg3,while red ginseng detected both s-type and r-type Rg3,and garden ginseng failed to detect Rg3.Conclusion HPLC method for the determination of ginsenoside Rg3 is sensitive,accurate and reproducible,suitable for the quantification of Rg3 in ginseng and its products.
