复发性流产患者吲哚胺2,3-双加氧酶及CD56、CD163表达分析 被引量:1

Analysis of the expression of indoleamine 2,3-dioxygenase, CD56 and CD163 in patients with recurrent spontaneous abortion

摘要 目的探讨复发性流产(RSA)患者吲哚胺2,3-双加氧酶(IDO)及CD56、CD163的表达分析。方法选取2020年2月至2021年4月解放军总医院第六医学中心诊治的128例RSA患者作为研究对象,将其设为研究组。另选取同期解放军总医院第六医学中心收取的128例正常妊娠女性设为对照组。采集两组的子宫内膜组织细胞进行培养观察,并定量测定样本中IDO、CD56以及CD163的表达水平。结果研究组IDO阳性表达率为46.09%,低于对照组的79.69%,差异具有统计学意义(P<0.05);对照组子宫内膜样本的免疫组化染色结果可观察到明显的IDO表达,研究组则以局部染色、弱染色为主。研究组CD56(+)、CD56^(+)表达水平明显高于对照组,差异具有统计学意义(P<0.05);两组CD163^(+)、CD163^(+)表达水平比较,差异无统计学意义(P>0.05)。CD56染色结果可见研究组存在局部CD56的明显阳性表达,但整体仍趋于弱染色,对照组CD56阳性表达并不明显,整体呈现弱染色。CD163染色结果在两组中均呈现明显的局部棕色,整体呈现中度染色,且两组表达并无明显差异。结论在RSA患者子宫内膜细胞中,IDO与CD56表达量呈上升趋势;RSA的发病机制可能与自身免疫存在密切关系,但是否与CD163的抑制性免疫调节机制失调相关,目前尚不清楚。 Objective To investigate the expression of indoleamine 2,3-dioxygenase(IDO), CD56 and CD163 in patients with recurrent spontaneous abortion(RSA). Methods 128 patients with RSA treated in the Sixth Medical Center, PLA General Hospital from February 2020 to April 2021 were selected as the study group. Another 128 normal pregnant women collected from the Sixth Medical Center, PLA General Hospital during the same period were selected as the control group. Endometrial tissue cells of the two groups were collected for culture observation, and the expression levels of IDO, CD56 and CD163 in the samples were quantitatively determined. Results The positive expression rate of IDO in the study group was 46.09%, lower than 79.69% in the control group(P<0.05). Immunohistochemical staining results of endometrial samples in the control group showed obvious IDO expression, while in the study group, local staining and weak staining were dominant. The expression levels of CD56;and CD56;in the study group were significantly higher than those in the control group(P<0.05). There was no significant difference in the expression levels of CD163^(+)and CD163^(+)between the two groups(P>0.05). CD56^(+) staining results showed that there was obvious positive expression of local CD56^(+)in the study group, but the overall staining was still weak, while the positive expression of CD56 in the control group was not obvious, and the overall staining was weak. CD163 staining results showed obvious local brown color in both groups and moderate staining in the whole, and there was no significant difference in expression between the two groups. Conclusions The expression levels of IDO and CD56 in endometrial cells of RSA patients showed an upward trend. The pathogenesis of RSA may be closely related to autoimmunity, but whether it is related to the dysregulation of inhibitory immune regulation of CD163 is not clear at present.
