间歇浸没液培技术在香蕉繁育中的应用探究

摘要 香蕉种苗繁育的组培方法主要是传统固体培养基培养,其劳动量大,增殖系数不高,自动化与机械化程度低,生产效益提升空间有限。本文利用气压式间歇浸没液体培养、固体静置培养、液体静置培养3种不同的培养方式组培研究香蕉种苗,探究在增殖率方面具有积极作用的香蕉组培技术。实验结果:(1)液体间歇浸没培养的香蕉无菌苗增殖率为3.55,是传统固体培养的1.12倍,是液体培养的1.73倍;(2)在液体间歇浸没培养香蕉无菌苗试验中,培养效果最好的间歇浸没频率为10 min/2 h;(3)液体间歇性浸没组培的苗长势好,生根较少,叶茎粗壮,分蘖多,优于传统组培方式。 The tissue culture method used in banana seedling breeding is mainly traditional solid medium culture,which has a large amount of labor,low proliferation coefficient,low degree of automation and mechanization,and limited space for improvement of production efficiency.In this study,three different culture methods were used to study the tissue culture of banana seedlings,namely,intermittent submerged liquid culture,solid standing culture and liquid standing culture,to explore the positive role of banana tissue culture technology in the proliferation rate.The experimental results are as follows:(1)The proliferation rate of sterile banana seedlings in liquid intermittent immersion culture was 3.55,1.12 times of that in traditional solid culture and 1.73 times of that in liquid culture;(2)In the experiment of liquid intermittent immersion culture of sterile banana seedlings,the frequency of intermittent immersion with the best culture effect was 10 min/2 h;(3)Liquid intermittent immersion tissue culture method of tissue culture seedlings grow better,less rooting,thick leaves and stems,more tillers,better than the traditional tissue culture method.
