摘要
目的针对槲皮素的抗癌功效,研究不同浓度槲皮素对胃癌细胞SGC-7901细胞增殖和凋亡的影响以及对p-PI3K信号蛋白和抗凋亡蛋白Bcl-2的作用。方法不同浓度的槲皮素(0、20、40、60、80、100μmol)干预细胞后,采用CCK-8法检测SCG-7901细胞增殖情况,采用hoechst 33342染色,观察细胞核变化,流式细胞凋亡分析槲皮素对细胞凋亡的影响,以及Western Blotting检测槲皮素对p-PI3K和Bcl-2蛋白表达的影响。结果槲皮素对胃癌细胞SGC-7901的活性具有抑制效应,且这种效应呈现浓度依赖性;荧光显微镜下观察到随着槲皮素浓度的增高,细胞核发生皱缩、核裂变,DNA出现片段化、染色质固缩、染色加深、染色质形态改变等现象。流式细胞凋亡实验证实槲皮素对胃癌SGC-7901具有诱导细胞凋亡的作用,Western Blotting实验发现槲皮素能抑制胃癌细胞SGC-7901内p-PI3K和Bcl-2蛋白的表达。结论上述结果表明槲皮素能抑制人胃癌SGC-7901细胞增殖并诱导其凋亡,并能调节p-PI3K和Bcl-2蛋白的表达,其可以作为抗胃癌细胞活性的药物进行药物靶点研究。
OBJECTIVE In order to clarify the anticancer efficacy of quercetin,studied the effects of different concentrations of quercetin on the proliferation and apoptosis of gastric cancer cell SGC-7901 as well as the effects on p-PI3K signaling protein and anti-apoptotic protein Bcl-2 were studied.METHODS SGC-7901 were treated with Different concentrations of quercetin(0、20、40、60、80、100μmol),CCK-8 detected the cell proliferation,nucleus was observed under fluorescence microscope by hochest 33342 stain,the effect of quercetin on cell apoptosis analyzed by flow cytometry,and the effect of quercetin on the expression of P-PI3K and Bcl-2 proteins detected by Western Blotting.RESULTS Quercetin had an inhibitory effect on the activity of Gastric cancer cell SGC-7901,which was concentration dependent.Fluorescence microscope showed that with the increase of quercetin concentration,the nucleus shrivel and nuclear fission occurred in the nucleus,DNA fragmentation,chromatin solidification,chromatin deepening and chromatin morphosis.Flow cytometry experiments confirmed that quercetin could induce apoptosis in gastric cancer cell line SGC-7901.Western Blotting experiments showed that quercetin could inhibit the expression of p-PI3K and Bcl-2 proteins.CONCLUSION The results indicated that quercetin could inhibit the proliferation and induce the apoptosis of human gastric cancer SGC-7901 cells,regulate the expression of p-PI3K and Bcl-2 proteins,and could be used as a drug for anti-gastric cancer cell activity research.
