红豆树化学成分及其抗炎活性研究

摘要 目的研究红豆树种子化学成分。方法采用聚酰胺、凝胶树脂等柱色谱和高效液相色谱、核磁共振色谱等分析手段对红豆树乙酸乙酯提取部位进行系统分离以及纯化。结果分离得到8个(异)黄酮及其苷类化合物,分别为:芒柄花苷(1),4′,8-二甲氧基-7-O-β-D-吡喃葡萄糖基异黄酮(2),圆荚草双糖苷(3),染料木素-7-O-β-D-呋喃芹糖基-(1→6)-O-β-D-吡喃葡萄糖苷(4),芦丁(5),山奈酚-3-O-芸香糖苷(6),水仙苷(7)和番石榴苷(8)。采用脂多糖(LPS)刺激小鼠单核巨噬细胞RAW264.7对单体化合物进行抗炎活性评价。结论化合物7和8为红豆属植物中首次分离得到,化合物1~6为首次从红豆树种子中分离得到。抗炎活性结果表明,在浓度25μmol·L^(-1)时,化合物1~8具有抑制炎症介质一氧化氮(NO)释放的效果,其中2~4和6抗炎效果较好。 OBJECTIVE To study the chemical constituents from seeds of Ormosia hosiei Hemsl.&E.H.Wilson.METHODS Chemical constituents were isolated by chromatographic methods and identified by NMR analysis.Using LPS-activated RAW 264.7 cell line models in vitro,main compounds were evaluated for the inhibition against NO production.RESULTS Eight flavonoids were isolated from ethyl acetate of seeds of Ormosia hosiei.Their structures were identified as:ononin(1),4′,8-dimethoxy-7-O-β-D-glucopyranosylisoflavone(2),sphaerobioside(3),ambocin(4),rutin(5),kaempferol-3-rutinoside(6),narcissin(7)and guaijaverin(8).Among them,compounds 7 and 8 were isolated from the plants of the genus Ormosia for the first time,compounds 1-6 were isolated from the seeds of Ormosia hosiei for the first time.And compounds 1-8 have anti-inflammatory effects,among them,compounds 2-4,and 6 show strong activity.
