miR-3607在胃癌中的表达及其与预后的相关性研究

Expression of miR-3607 in gastric cancer and its correlation with prognosis

目的探讨miR-3607异常表达在胃癌细胞增殖、迁移和侵袭中的作用及其与患者预后的相关性。方法选择我院行胃癌切除术的患者117例,经手术切除获取胃癌组织和相邻正常组织(癌旁组织)。将miR-3607 mimic、miR-3607 inhibitor、mimic NC、inhibitor NC转染至HGC-27和MKN45细胞内(未处理细胞为Control)。采用实时定量PCR技术检测miR-3607的表达情况。CCK-8法检测胃癌细胞增殖情况。用Transwell法检测胃癌细胞的侵袭和迁移能力。采用Kaplan-Meier曲线和Cox回归分析探讨miR-3607的表达水平在胃癌中的预后意义。结果miR-3607在胃癌组织和胃癌细胞系中表达明显下调。miR-3607低表达可促进胃癌细胞的增殖、迁移和侵袭。Kaplan-Meier曲线显示,miR-3607低表达组患者总生存率显著低于高表达组。Cox回归分析结果显示,miR-3607低表达、淋巴结转移、TNMⅢ期与胃癌患者的总生存率具有相关性。结论miR-3607的异常低表达会促进胃癌的进展,并且通常提示患者的不良预后,miR-3607可能是胃癌的一个预后标志物和胃癌靶点治疗新策略。

Objective To investigate the effect of abnormal expression of miR-3607 on the proliferation,migration and invasion of gastric cancer cells and its correlation with the prognosis of patients.Methods A total of 117 patients who underwent gastric cancer resection in our hospital were selected,and the gastric cancer tissues and adjacent normal tissues(para-cancer tissues)were obtained after surgical resection.MiR-3607 mimic,miR-3607 inhibitor,mimic NC,inhibitor NC were transfected into the HGC-27 and MKN45 cells(the untreated cells were Control).The real-time quantitative PCR technology was used to detect the expression of miR-3607.CCK-8 method was used to detect the proliferation of gastric cancer cells.Transwell assay was used to detect the invasion and migration ability of gastric cancer cells.The Kaplan-Meier curve and Cox regression analysis were used to explore the prognostic significance of the expression level of miR-3607 in gastric cancer.Results The expression of miR-3607 was significantly down-regulated in gastric cancer tissues and gastric cancer cell lines.The low expression of miR-3607 can promote the proliferation,migration,and invasion of gastric cancer cells.The Kaplan-Meier curve showed that the overall survival rate of patients in the low-expression group of miR-3607 was significantly lower than that in the high-expression group.The results of Cox regression analysis showed that the low expression of miR-3607,lymph node metastasis,and TNM stageⅢhad correlation with the overall survival rate of gastric cancer patients.Conclusion The abnormally low expression of miR-3607 can promote the progression of gastric cancer and generally indicates a poor prognosis for patients,which may be a prognostic marker of gastric cancer and a new therapeutic strategy for targeted treatment of gastric cancer.
